摘要 目的:探析小儿厌食症模型大鼠血清代谢组特征,从代谢组角度探析小儿厌食症生物学本质。方法:采用基于液相色谱质谱联用的代谢组学技术检测小儿厌食症模型大鼠血清代谢物变化,筛选出小儿厌食症模型代谢差异物,分析其代谢通路及生物学意义。结果:模型组大鼠血清代谢物与空白组有显著差异,经数据处理,筛选出鞘氨醇、谷氨酸、柠檬酸等30个代谢差异物;涉及脂类代谢、氨基酸代谢及三羧酸循环等14条代谢通路。结论:模型组大鼠经特制高脂饲料喂养后产生病理性代谢产物,可引起脂质代谢、氨基酸代谢和能量代谢的紊乱,最终导致厌食症的发生;此外,还发现小儿厌食症和肠道菌群失调有很大相关,还需进一步探讨研究。 Objective:To analyze the characteristics of serum metabolic changes in model rats with infantile anorexia,and to explain the biological essence of anorexia in children from the perspective of metabolomics.Methods:Metabolomics technology based on liquid chromatography-mass spectrometry(LC-MS)was used to detect the changes of serum metabolites in model rats with infantile anorexia and to screen out metabolic differences to analyze its metabolic pathways and biological significance.Results:Serum metabolites in the model group were significantly different from the blank group.After data processing,30 different metabolites were screened out,such as sphingosine,glutamic acid and citric acid,which were involved in 14 metabolic pathways,such as lipid metabolism,amino acid metabolism and Krebs cycle.Conclusion:The differences in the metabolic profiles between the model group and the blank group suggest that the disorders of the metabolic network in infantile anorexia model are mainly manifested as disorder and attenuation of lipid metabolism,amino acid metabolism and energy metabolism.The incidence of infantile anorexia may be related to alteration of intestinal flora.
