冠豸山铁皮石斛种质资源遗传多样性分析 被引量:6

Genetic Diversity Analysis of Dendrobium officinale Germplasm Resources in Guanzhaishan

摘要 为了分析研究连城县冠豸山野生铁皮石斛种质资源的遗传多样性,为铁皮石斛种质资源的保护与杂交选育优良品种提供依据。采用农艺综合性状及SSR标记技术对12份冠豸山铁皮石斛供试材料进行种质多样性分析,同时对种质之间进行人工杂交授粉结实研究。筛选出6对多态性较好的引物,每对SSR引物扩增出等位位点数为2~5个,共扩增出20个等位位点,多态性占比为73.6%,聚类分析结果显示,供试的12份冠豸山铁皮石斛种质资源可分为3个类群;通过形态特征特性观察研究,连城县冠豸山野生铁皮石斛种质资源可分为五大类;种质资源间人工杂交授粉结实率为55%~100%,杂交与自交的授粉结实率存在显著差异。结果表明,冠豸山野生铁皮石斛种质资源遗传性状丰富,利用形态特征结合SSR标记技术种质资源多样性分析,指导选择亲缘关系较远的父、母本进行杂交育种,可提高育种效率。 The aim of this study was to investigate and analyze the genetic diversity of wild Dendrobium officinale germplasm reso urces from Guanzhaishan of Liancheng County, so as to provide a basis for the protection of germplasm resources of Dendrobium officinale and breeding of fine varieties. The germplasm diversity of 12 samples of Dendrobium officinale was analyzed by comprehensive agronomic characteristics and simple sequence repeat(SSR) marker technique. Also, studies on artificial cross pollination and fruiting of germplasm were carried out at the same time. 6 pairs of primers with good polymorphism were screened out. Each pair of SSR primers amplified 2~5 alleles, and 20 alleles were amplified. The percentage of polymorphism was 73.6%. The results of cluster analysis showed that 12 Dendrobium officinale germplasm resources from Guanzhaishan could be divided into3 groups. In addition, the wild Dendrobium officinale germplasm from Guanzhaishan of Liancheng County could be divided into five categories through morphological observation. The seed setting rate of artificial crossing pollination among germplasm resources was 55%~100%, and there was a significant difference in pollination and seed setting rates between hybridization and selfing. This study indicated that the wild Dendrobium officinale germplasm was rich in genetic character. Combining morphological characteristics with SSR marker technique to analyze the diversity of germplasm resources and guide the selection of distant male and female parents for cross breeding could improve the breeding efficiency.
