摘要
为了探讨山慈菇正丁醇提取物对NR8383细胞(大鼠肺泡巨噬细胞)分泌的白介素-1β(interleukin-1,IL-1β)和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平的影响,通过实时荧光定量PCR(real-time quantitative PCR,RT-qPCR)法检测TNF-α和IL-1β的mRNA水平,利用酶联免疫吸附法(enzyme-linked immune sorbent assay,ELISA)检测细胞上清液中TNF-α和IL-1β的蛋白含量。结果显示:与空白对照组相比,终浓度1.25 mg/mL的山慈菇正丁醇提取物可极显著降低R8383细胞分泌TNF-αmRNA水平和蛋白含量(P<0.01);与空白对照组相比,终浓度1.25 mg/mL山慈菇正丁醇提取物能极显著升高IL-1β的mRNA水平和蛋白含量(P<0.01),同时0.012 5 mg/mL山慈菇正丁醇提取物能极显著降低IL-1β的mRNA水平和蛋白含量(P<0.01)。说明终浓度为1.25 mg/mL的山慈菇正丁醇提取物可极显著调控NR8383细胞分泌的TNF-α和IL-1β的基因和蛋白含量。
In order to study the effects of n-butanol extract from Cremastra appendiculata on the levels of il-1β(interleukin-1,IL-1β)and TNF-α(tumor necrosis factor-α,TNF-α)from NR8383 cells(rat alveolar macrophages).Real-time quantitative PCR(RT-qPCR)and enzyme-linked immune sorbent assay(ELISA)were used to determinate the contents of TNF-αand il-1βin the supernatants of NR8383 cells.The results show that compared with the control group,the final concentration of 1.25 mg/mL n-butanol extract from Cremastra appendiculata could significantly decrease the secretion of NR8383 cells TNF-αmRNA level and protein content(P<0.01).Compared with the control group,the final concentration of 1.25 mg/mL n-butanol extract from Cremastra appendiculata could significantly increase the secretion of NR8383 cells IL-1βmRNA level and protein content(P<0.01),while 0.012 5 mg/mL n-butanol extract from Cremastra appendiculata could significantly decrease the secretion of NR8383 cells IL-1βmRNA level and protein content(P<0.01).It is concluded that the final concentration of 1.25 mg/mL n-butanol extract from Cremastra appendiculata could significantly regulate the gene and protein contents of TNF-αand IL-1βsecreted by NR8383 cells.
