摘要
目的探讨加巴喷丁对脊神经结扎(SNL)大鼠神经病理性疼痛的作用及其机制。方法将36只雌性SD大鼠随机均分为3组:脊神经结扎组(SNL组,行脊神经结扎手术)、加巴喷丁组[GBP组,行脊神经结扎手术并腹腔注射加巴喷丁(50 mg/kg)],假手术组(Sham组,不结扎脊神经,其他手术步骤与SNL组一致),每组12只。分别在术前,术后1、3、5、7 d检测各组大鼠热缩足潜伏期(TWL)和机械缩足反应阈值(MWT);Western blotting检测大鼠术后7 d鼠背根神经节中Toll样受体4(TLR4)表达。结果与Sham组比较,SNL组、GBP组在术后各时间点MWT和TWL均降低(均P<0.05);术后5、7 d GBP组MWT和TWL均高于SNL组(均P<0.05)。与Sham组比较,SNL组大鼠背根神经节中TLR4表达明显升高(P<0.05);而GBP组大鼠背根神经节中TLR4表达升高不明显(P>0.05)。与SNL组比较,GBP组大鼠背根神经节中TLR4表达明显下降(P<0.05)。结论腹腔注射加巴喷丁可缓解大鼠脊神经结扎导致的神经疼痛,下调大鼠背根神经节中TLR4表达,加巴喷丁对神经病理性疼痛具有镇痛作用。
Objective To investigate the analgesic effect of gabapentin(GBP)on neuropathic pain in rats and its effect on the expres-sion of Toll-like receptor 4(TLR4)in the dorsal root ganglion(DRG)after spinal nerve ligation.Methods Thirty-six female SD rats were randomly divided into three groups:the spinal nerve ligation(SNL)group(n=12);GBP group(n=12);and sham group(n=12).The thermal withdrawal latency(TWL)and mechanical withdrawal threshold(MWT)of rats in each group were measured at preoperative(Day 0)and postoperative(Days 1,3,5,and 7)time points.The changes in TLR4 in the DRG of rats on Day 7 after SNL were detected by Western blotting.Results At each postoperative time point,the MWT and TWL levels were lower in the SNL and GBP groups(P<0.05)than in the sham group and reached the lowest level on Day 7 after the operation(P<0.05).Levels were higher in the GBP group than in the SNL group on Days 5 and 7(P<0.05).On Day 7,the protein levels of TLR4 in the DRG of the SNL group were significantly increased(P<0.05)and the levels were lower in the GBP group than in the SNL group.Conclusion Intraperitoneal injection of GBP can relieve neuropathic pain caused by nerve ligation in rats and downregulate the expression of TLR4 in the DRG and has an analgesic effect on neuropathic pain.
