摘要
目的:探讨程序性细胞死亡因子4(PDCD4)过表达对增生性瘢痕成纤维细胞增殖及凋亡的影响。方法:分离培养增生性瘢痕成纤维细胞,分别转染pEGFP-PDCD4、pEGFP,以未转染细胞为空白对照,采用qRT-PCR和Western blot法检测细胞中PDCD4水平确定转染效果。MTT法检测3组细胞增殖活性,TUNEL法检测细胞凋亡,Western blot法检测PTEN、Bax、Bcl-2表达水平。结果:与空白对照组和pEGFP组相比,pEGFP-PDCD4组细胞中PDCD4表达水平升高;细胞增殖活性降低,细胞凋亡率升高,同时细胞中PTEN、Bax表达水平升高,Bcl-2表达水平下降(P<0.05)。结论:过表达PDCD4可抑制增生性瘢痕成纤维细胞增殖,促进细胞凋亡,其机制可能与上调细胞中PTEN、Bax的表达,下调Bcl-2的表达有关。
Aim:To investigate the effects of programmed cell death 4(PDCD4)overexpression on proliferation and apoptosis of hyperplastic scar fibroblasts.Methods:The hyperplastic scar fibroblasts were cultured and transfected with pEGFP-PDCD4,pEGFP,and the cells not transfected were blank control.qRT-PCR and Western blot were used to detect the level of PDCD4 in cells,MTT assay was used to detect cell proliferation,TUNEL assay was used to detect apoptosis,and Western blot was used to detect the levels of PTEN,Bax and Bcl-2 protein.Results:Compared with blank control group and pEGFP group,the expression of PDCD4 level in pEGFP-PDCD4 group was increased;cell proliferation capability decreased,apoptosis rate increased,the expression levels of PTEN and Bax in the cells were increased,and the level of Bcl-2 in the cells was decreased(P<0.05).Conclusion:Overexpression of PDCD4 inhibits the proliferation and promote the apoptosis of hyperplastic scar fibroblasts.The mechanism may be related to up-regulation of PTEN and Bax expressions and down-regulation of Bcl-2 expression in cells.
