犀角散对肝衰竭小鼠肝脏固有免疫的影响 被引量:2

摘要 目的:通过观察犀角散对肝衰竭模型小鼠肝脏固有免疫的作用,探讨犀角散治疗肝衰竭的可能途径。方法:将60只小鼠按数字表法随机分为犀角散组(A组)、对照组(B组)、生理盐水组(C组),正常小鼠组(D组),并分别灌胃给予干预处理,A、B、C 3组小鼠运用卡介苗+脂多糖建成肝衰竭小鼠模型,D组不做造模处理;13 d后,观察各组肝脏结构、肝内NLRP3、caspase-1、白细胞介素(IL)-1β蛋白及基因表达。结果:A、B、C 3组小鼠都可见到肝组织中明显的肝细胞坏死,A组小鼠肝组织中肝细胞坏死程度较轻;3组造模肝衰竭小鼠肝组织中NLRP3、caspase-1、IL-1β蛋白及基因表达水平较D组小鼠明显升高;犀角散组小鼠下调肝组织NLRP3、caspase-1蛋白及基因表达水平。结论:肝衰竭小鼠肝组织中存在NLRP3炎症复合物通路激活,犀角散可以通过影响NLRP3炎症复合物通路达到阻断肝脏炎症反应的目的。 Objective:To investigate the effects of Xijiaosan Recipe on innate immune of liver failure mice.Methods:Sixty mice were randomly divided into 4 groups:Xijiaosan Recipe group(A group);control group(B group);NS group(C group);normal group(D group).A,B,C group were induced by Bacille Calmette-Guerin and lipopolysaccharide.The mice were sacrificed on the 13th day.The liver tissue of mice were measured.The expression of NLRP3,caspase-1,IL-1β protein and mRNA in liver issue were detected.Results:Subsequent LPS challenge in BCG-primed mice induced multilobular hepatocellular necrosis in liver tissue.The hepatocellular necrosis of Xijiaosan Recipe group was lighter.The levels of NLRP3,caspase-1,IL-1βprotein and mRNA were significantly increased in the liver failure mice s liver issue.Xijiaosan Recipe down-regulated the levels of NLRP3,caspase-1,IL-1βprotein and mRNA in the liver failure mice s liver issue.Conclusion:The NLRP3 inflammasome signaling pathway was activity in the liver failure mice s liver issue.The anti-inflammatory mechanism of Xijiaosan Recipe may be related to decreasing the expression levels of the NLRP3 inflammasome signaling pathway.
