中间锦鸡儿CibHLH93基因克隆及表达分析

摘要 b HLH属于转录调控因子中的一个大家族,广泛存在于真核生物界,因具有保守性较高的b HLH结构域而得名。b HLH以二聚体形式特异性结合于DNA靶位点,调控多样化的生物过程,对真核生物的生长发育和次生代谢必不可少。植物中,b HLH转录因子通过与其他转录因子互作激活胁迫基因的表达,从而使植物免受非生物胁迫带来的伤害。本研究从实验室干旱转录组数据库中,得到一条中间锦鸡儿b HLH基因全长序列。序列分析显示该基因的开放阅读框为999 bp,编码332个氨基酸。比对分析结果发现其推导的氨基酸序列具有b HLH保守结构域,与拟南芥b HLH93蛋白一致性为68%,将其命名为Cib HLH93。实时荧光定量PCR结果显示,Cib HLH93受到Na Cl、冷、热和脱水的诱导,表达量随处理时间延长呈下降趋势,推测该基因在响应非生物胁迫过程中是一个下调基因。拟南芥叶肉原生质体瞬时表达实验表明,Cib HLH93主要定位于细胞核。以上结果,为进一步揭示Cib HLH93功能的研究提供了基础。 Basic/helix-loop-helix(b HLH) transcription factors(TFs) belong to a super family of transcriptional regulators, which widely exist in eukaryotic kingdoms, and it were named for its highly conserved of b HLH domain. b HLH regulates the diverse biological processes by specifically binding to the target DNA as dimers, it is essential for the development and secondary metabolism of eukaryotes. b HLH can interact with other transcription factors to activate the expression of stress genes to protect plants from damage caused by abiotic stress. In this research, a b HLH gene with full length sequence from Caragana intermedia was isolated from dehydration treated transcriptome in our lab. Sequence analysis showed that this gene has an open reading frame of 999 bp which encoded a protein composed of 332 amino acids. Blast analysis indicated that the deduced amino acid sequences included a conserved b HLH domain, and shared 68% identities with Atb HLH93 protein, so it was named as Cib HLH93. Real-time quantitative PCR showed that Cib HLH93 was induced by Na Cl, cold, hot and dehydration treatments, and the expression level was decreased with the proceeding of treatments, indicating that the gene was a down-regulated gene in responding to abiotic stress. Transient expression assay revealed that Cib HLH93 localized in nuclei in Arabidopsis mesophyll protoplast. Taken together these results laid foundation for understanding the function of Cib HLH93.
