应用SSR和SRAP分析苦瓜育种新组合的亲缘关系 被引量:6

Analysis on Genetic Relationship of New Breeding Combinations Based on SSR and SRAP Markers in Momordica charantia L.

摘要 苦瓜遗传背景复杂,采用分子标记技术分析和评价苦瓜育种新组合的亲缘关系,对苦瓜优良品种的选育至关重要。本研究利用SSR标记和SRAP标记对22份苦瓜育种新组合材料进行了亲缘关系的分析,结果显示,筛选出的8对SSR和l0对SRAP特异引物的扩增结果均具有较高的多态性信息含量,平均多态性百分率为72.71%和83.80%。聚类分析结果发现,SSR和SRAP标记均能有效揭示苦瓜的遗传多样性,2种标记的聚类结果一致性高,在GS为0.71和0.64时,SSR和SRAP均可将22份材料分为4类。2种标记综合能够更准确地分析22份材料的亲缘关系。在GS为0.66时,22份材料被分为5类,在2种标记聚类分析中,亲缘关系较远的BL组合、YS31、JC21和MY023都各自聚类,HY、GL和ZY组合又聚为一类,这与GL组合和ZY组合是HY组合的父母本的事实相吻合。本研究结果表明,采用2种标记对苦瓜育种新组合进行综合分析,能更准确地反映苦瓜材料间的亲缘关系。 Genetic relationship analysis of new breeding combinations based on molecular marker technology is essential to screen improved variety in Momordica charantia because of its complicated genetic background. 22 new breeding materials were analyzed based on SSR and SRAP in this paper. 8 pairs of SSR primers and 10 pairs of SRAP primers were screened with high PIC, average polymorphic percentage achieved 72.71% and 83.80%separately in SSR and SRAP amplification results. Cluster analysis showed that both SSR and SRAP could display genetic diversity of all materials efficiently, 2 kind of marker result showed high identity, all materials were divided into 4 classes according to GS value of 0.71 in SSR and 0.64 in SRAP. Complicated 2 markers could uncover the genetic relationship of 22 materials more accurately, all materials were divided in to 5 classes according to GS value of 0.76. BL combination, YS31, JC21 and MY023, farer genetic relationship displayed by SSR and SRAP cluster analysis individually, clustered separately in complicated cluster analysis. 3 groups of HY, GL and ZY combination all clustered into 1 class again, it fit to the fact that HY combination was hybrid from GL and ZY combinations. Results indicated that complicated 2 markers analysis could uncover materials' genetic relationship more accurately in Momordica charantia.
