摘要
目的 观察维甲酸 (RA)作用人胃癌细胞系GHC - 2 7后 ,细胞增殖、细胞周期及细胞周期素依赖性激酶抑制蛋白 (CKIs) (P16、P2 1、P2 7)表达的变化 ,探讨RA抑制HGC 2 7增殖 ,促进其分化的作用机制。方法 不同浓度RA作用HGC 2 7细胞后 ,MTT法检测细胞增殖情况 ,流式细胞仪检测细胞周期 ,Westermblot、Northernblot分别检测CKIs(P16、P2 1、P2 7)蛋白、mRNA水平。结果 在一定浓度范围之内 ,RA能抑制HGC 2 7细胞增殖 ,细胞周期发生G1→S期阻滞 ,并上调P2 1、P2 7的蛋白、mRNA水平 ,但对P16的蛋白、mRNA水平没有影响。结论 RA能通过上调CKIs尤其是P2 1、P2 7的表达而抑制HGC 2 7增殖 ,促进其分化。
Purpose: To study the proliferation, cell cycle and expression of cyclindependent kinase inhibitors (CKIs) of human gastric carcinoma cell line HGC-27 treated with retinoic acid (RA), and to investigate the mechanism of RA to inhibit tumor cell's growth and induce their differentiation. Methods: We treated HGC-27 cells with varible dose RA, detected the proliferation by MTT method and observed cell cycle by flow cytometer, used Western blot as well as Northern blot to detect the protein and mRNA level of CKIs (P16, P21, P27) respectively. Results: RA could inhibit the proliferation of HGC-27, bring about G1 phase arrest and up-regulate the level of protein and mRNA of P21 as well as P27, but has no effect on the level of protein and mRNA of P16. Conclusions: RA could inhibit proliferation and induce differentiation on HGC-27 cells by up-regulating experession of CKIs, especially P21 and p27.
