摘要
目的 观察视黄酸对人横纹肌肉瘤细胞的诱导分化作用及对生肌调节因子 (MRF)表达的影响。方法 以 1 0 -6mol/L全反式维甲酸 (ATRA)处理人横纹肌肉瘤细胞RD和A6 73,用免疫细胞化学检测横纹肌肌球蛋白重链和肌动蛋白α actin表达 ,用原位杂交检测生肌调节因子MRF4、MyoDmRNA表达 ,并观察细胞生长曲线变化。结果 ATRA作用后 ,RD、A6 73细胞肌球蛋白重链和α actin表达增强 ,MRF4和MyoDmRNA表达上调 ,细胞生长受抑。结论 ATRA具有促进横纹肌肉瘤细胞分化的作用 ,其作用可能与MRF表达上调有关。
Objective To study the effects of retinoic acid on the differentiation of human rhabdomyosarcoma cells and on the expression of myogenic regulatory factor (MRF). Methods Human rhabdomyosarcoma cell line RD and A673 were treated with 10 -6 mol/L all tans retinoic acid (ATRA). The expressions of myosin heavy chain (MHC) and α action in the cells were determined with immunocytochemical method and the expression of myogenic regulatory factors MRF4 and MyoD mRNA were studied with in situ hybridization. The cell growth curve was observed at the same time. Results The expression of MHC and α action were increased in RD and A673 cells after treated with ATRA, while the expressions of MRF4 and MyoD mRNA were up regulated and cell growth was suppressed. Conclusion ATRA can induce the differentiation of RD and A673 cells and inhibit the cell growth in vitro , which might be associated with the up regulated expression of MRF.
