摘要
多元药物抗性基因(multidrug resistance gene,mdr-1)编码 P 170膜蛋白,可使细胞对多种结构不同的细胞毒药物产生抗性。本文以 mdr-1 cDNA为探针,做白细胞总 RNA 斑点杂交分析了5例髓性白血病人mdr-1的mRNA水平。说明mdr-1基因在此类患者的白细胞中的表达有明显差异,同一病人治疗前后也有改变。所建立的观查临床标本中基因表达水平的方法,具广泛的应用价值。
P-glycoprotein (Pgp; Mr=170,000) is encoded by a family of genes-Multidrug resistance gene.The Pgp bas been demonstrated to mediate resistance to multiple structurally dissimilar drugs, whichfuctions as an energy-dependent efflux pump so that a cell with high level of mdr expression canmore effectively eliminate cytotoxic drugs. In this report, a simplified method for analysis of clinicalsamples and assess the level of gene expression was set up. Furthermore, by using ^(32)P labelled mdr-1cDNA as the probe and RNA dot blotting the mdr-mRNAs from 5 cases of myeloblastic leukemiacells were analysed. It was shown that the level of mdr-1 expression in different myeloleukemic cellswas various and reduced in one case after remission. The established method for mRNA analysis couldbe generalized for evaluating the level of mRNA in clinical samples.
