摘要
目的 探讨多药耐药 (MDR)基因产物在乳腺癌组织中的表达及其相互关系。方法 应用免疫组织化学方法 ,检测6 9例乳腺癌组织中MDR基因产物 (P gp、GST π、TOPOⅡ )。结果 乳腺癌组织中以P gp和GST π表达为主 ,其表达率分别为88 4 1%和 79 71% ,二者均明显高于TOPOⅡ表达率 5 7 97% (P <0 0 1) ;2种MDR基因产物共表达率为 33 33% ,3种MDR基因产物共表达率为 5 5 0 7% ,均明显高于单独表达率 11 5 9% (P <0 0 1) ;GST π和TOPOⅡ的表达均同时伴有P gp的表达 ,在P gp为阴性情况下 ,没有发现GST π和TOPOⅡ的单独表达和共表达现象。P gp和GST π的表达呈显著正相关 (P <0 0 1) ;P gp和TOPOⅡ的表达呈明显负相关 (P <0 0 5 ) ;GST π和TOPOⅡ的表达呈明显负相关 (P <0 0 5 )。结论 P gp、GST π、TOPOⅡ在乳腺癌组织中表达对肿瘤耐药起重要作用 ,单基因和多基因协同作用 ,以多基因共表达为主。
Objective To explore expression of multidrug resistance(MDR) gene production in breast cancer tissue.Methods A total of 69 case of paraffin embedded breast cancer tissue was stained immunohistochemically for P gp?GST π?TOPOⅡ.Results The positive expression of P gp and GST π separately were 88.41% and 79.71%, which were much higher than the positive expression of TOPOⅡ 57.97% in breast tissue;Among them, the coexpression rate of only 2 MDR gene production and only 2 or 3 MDR gene production was each 33.33% and 88.40%, which exceeded apparently more than individal expression rate 11.59%; The positive expression of GST π and TOPOⅡ often accompanied the expression of P gp;If the expression of P gp was negative, there was no the coexpression of GST π and TOPOⅡ.P gp significantly positively correlated with GST π;P gp negatively related to TOPOⅡ;GST π had obviously negative correlation with TOPOⅡ.Conclusion The expression of P gp?GST π?TOPOⅡ in breast cancer tissue made it important to influence tumor resistance,signal cooperated with many genes,but the coexpression of many gene mostly.
