摘要
目的:建立小剂量胺碘酮(AD)治疗心律失常时固相萃取-高效液相色谱血清浓度测定方法。方法:用Waters OA-SIS^(TM) HLB小柱预处理样品,70%的甲醇洗脱杂质,氯仿洗脱药物;采用ODS-C_8柱,流动相为甲醇-水-乙腈-四甲基乙二胺-冰醋酸(100:65:100:1:0.8),流速1.2 mL·min^(-1),柱温为45℃,二极管阵列检测器检测波长243 nm。结果:血清中AD在0.1~3.2mg·L^(-1)浓度范围内具有良好的线性关系,最低检测药物浓度为40ng·mL^(-1),日内与日间加样回收率分别为98.5%~101.3%与97.7%~99.5%(n=5),日内与日间HSD均低于5%(n=5)。结论:本方法简单、灵敏、准确、干扰小,适用于生物样品中AD的血药浓度监测。
Objective: To establish a SPE - HPLC method for the determination of amiodarone in human serum with low dose treating arrhythmia. Method: Amiodarone in serum was enriched with OSASIS?HLB Sep - Pak solid phase extraction column, purified by 70% methanol and extracted by chloroform. Using ODS -C18 column, the mobile phase consisted of methanol - water - acetonitrile - tetraethyl amine - acetic acid ( 100: 65 : 100: 1: 0. 8 ) , at a flow rate of 1. 2 mL · min-1. The column temperature was 45 ℃ and DAD detection wavelength was 243 nm. Results: The minimal detectable serum drug concentration was 40 ng · mL-1. The calibration curve was linear in the range of 0. 1-3. 2 mg · L-1 (r = 0. 999 6). The intra - day and inter - day recovery was 98. 5% -101. 3% and 97. 7% -99. 5% (n =5). Intra - day and inter - day RSD were less than 5% (n =5). Conclusion: The method is simple, sensitive and accurate. It is suitable for the determination of amiodarone in serum.
