摘要
采用高效均质酶解技术优化提取五味子多糖;采用单因素实验确定影响试验的关键因素和范围,Box-Behnken-Design试验、响应面优化试验模型和提取时间、提取温度和均质转速的最优提取条件。优化得到最佳提取条件为:果胶酶量为1.33%,提取时间1.92min,提取温度50.82℃,均质转速16319.73r/min,在此条件下多糖提取得率为10.03%,与模型的预测得率接近(9.39%),表明建立的模型适合优化多糖的提取条件。结果表明均质酶解是一种高效、环保的植物多糖提取技术,这些将会为多糖在健康食品和医药的广泛应用提供理论依据。
In this paper,an efficient homogenate enzymolysis extraction (HEE) technique was developed and optimized for the extraction of polysaccharide from Schisandra chinensis Baill.The single factor experiments were used to determine the key experimental factors and their test range.The response surface methodology (RSM) with a Box–Behnken Design (BBD) was employed to model and optimize three experimental operating conditions which are extraction time,extraction temperature and homogenizer speed.The optimized conditions were as follows: amount of pectinase 1.33%(wt.),extraction time 1.92 min,extraction temperature 50.82℃ and homogenizer speed 16319.73 r/min.Under these conditions,the maximal yield of crude polysaccharides was 10.03%,which is consistent with the predicted yield (9.39%),indicating the models are suitability for optimizing the polysaccharides extraction conditions.The results suggest that HEE could be an efficient and environment-friendly technique for the extraction of polysaccharide from plant.These may further provide theoretical basis for the widely application of polysaccharides in medicine and health care products.
