摘要
为研究能够同时检测PEDV、TGEV、PoRV三种猪病毒性腹泻病原的方法,分别设计三套特异性的引物和探针,建立了基于Taqman探针的三重Real-Time RT-PCR检测方法,实验结果表明,该方法特异性好,灵敏度高,检测最低浓度为10 copies/μL数量级.应用该方法对广东11个地市的44份猪腹泻样品进行检测,其中PEDV阳性率为100%,TGEV、PoRV阳性率为0,证明PEDV是引起广东地区猪病毒性腹泻的重要病原.
detect the porcine epidemic diarrhea virus(PEDV),transmissible gastroenteritis virus(TGEV)and porcine rotavirus(PoRV)simultaneously,three sets of specific primers and probes were designed and a triple real-time reverse transcription-polymerase chain reaction method based on TaqMan^TM probes was established. This method had good specificity and high sensitivity,and the minimum concentration was 10 copies/μL. Forty-four samples of porcine diarrhea were detected in 11 cities of Guangdong Province,China,with this method. The positive rate of PEDV was 100%,while both TGEV and PoRV were not detected. These data demonstrated that PEDV is an important pathogen causing porcine viral diarrhea in Guangdong Province.
