聚六亚甲基双胍缓释抗菌凝胶的制备和质量评价 被引量:3

摘要 〔目的〕在PHMB@ACPs的制备基础上,设计并优化抗菌凝胶处方,使之达到最优抗菌效果和缓释效果。〔方法〕利用已制备出的PHMB@ACPs颗粒通过单因素实验选出最优基质。再采用四因素三水平正交设计法以黏度和感官评分为依据优选出辅料用量。然后通过单因素抗菌实验确定凝胶剂的最强抗菌浓度。最后对优选出来的处方进行缓释性能、稳定性能、稀释后抗菌性能评价。〔结果〕成功筛选出最优处方:羟丙甲基纤维素150mg/g,甘油0.1mL/g,玫瑰水合物0.2mL/g,芦荟0.02g/g,PHMB@ACPs的用量为1.8mg/g。质量评价检测结果表明该凝胶具有缓释性能、良好的离心稳定性和优良的耐热耐寒性,且其pH测定值也符合人体腔道的弱酸环境。〔结论〕优选出的PHMB@ACPs芳香型抗菌凝胶剂与常见抗菌凝胶剂相比,缓释性和稳定性明显提高,药物的顺应性也明显改善。 [Objective]Based on the preparation of PHMB@ACPs, the antibacterial gel formulation was designed and optimized to achieve optimal antibacterial and sustained release effects.〔Methods〕Using the prepared PHMB@ACPs particles,we selected the optimal matrix by single factor experiments. Next we optimized the amount of excipients based on viscosity and sensory score by the four-factor three-level orthogonal design method. Then we confirmed the strongest antimicrobial concentration of the gel by a single factor antimicrobial assay. Finally, the preferred formulation was evaluated for sustained release performance, stability, and post-dilution antimicrobial performance.〔Results〕The optimal prescription was successfully screened: hydroxypropylmethylcellulose 150 mg/g, glycerol 0.1 mL/g, rose hydrate 0.2 mL/g, aloe vera 0.02 g/g, and PHMB@ACPs 1.8 mg/g. The results of quality evaluation test of the antibacterial gel showed that the gel containing PHMB@ ACPs has sustained release properties, excellent centrifugal stability, thermotolerance and cold tolerance. Its pH measurement also conforms to the environment of the human body.〔Conclusion〕Comparing to the conventional antibacterial gel,the preferred PHMB@ACPs aromatic antibacterial gel has markedly improvements not only of sustained release and stability,but also of the drug compliance.
