摘要
支架蛋白是细胞中的重要辅助分子,可通过与多种信号酶、受体或离子通道结合,招募各种信号分子影响细胞信号转导。虽然支架蛋白通常缺乏酶催化活性,但它们对控制信号信息流动仍有很大影响。近年来研究发现,有些支架蛋白可作为具有催化活性的酶发挥作用,而有些酶则可作为支架蛋白通过非酶活性发挥功能,进而参与不同的细胞信号转导过程。本文将对支架蛋白的特征、作用机制,具有酶催化活性支架蛋白的发现以及支架蛋白在细胞信号转导、物质代谢调控中的作用进行综述。
Scaffold proteins,as important accessory molecules in cells,affect cell signal transduction by binding to a variety of signaling enzymes,receptors or ion channels and recruiting different molecular signals. Although scaffold proteins generally lack catalytic activity,they still have a great impact on controlling the flow of signal information.It was discovered recently that a few scaffold proteins have enzymatic activities, while some enzymes could act as scaffold proteins independent of their enzyme activities.This paper reviewed the features and mechanisms of scaffold proteins,and discussed some recently discovered scaffold proteins that possess enzymatic activities.The effects of scaffold proteins on cellular signal transduction and metabolic regulation are also included.
