摘要
为了研究氯虫苯甲酰胺和甲氨基阿维菌素苯甲酸盐(甲维盐)对草地贪夜蛾Spodoptera frugiperda的毒杀作用及解毒机制,采用叶片浸渍法测定了2种杀虫剂对草地贪夜蛾3龄幼虫的LC10、LC20与LC50,并研究这2种农药的亚致死剂量(LC20)对幼虫的羧酸酯酶(CarE)、谷胱甘肽-S-转移酶(GST)、细胞色素P450(CYP450)3种主要解毒酶活性的影响。结果表明甲维盐对3龄幼虫的毒力更高,48h时LC10、LC20、LC50分别为0.005、0.007、0.013mg/L。2种农药处理72h内,幼虫的3种酶活力呈现不同的动态变化。氯虫苯甲酰胺对CarE活力表现先诱导后抑制的作用,在24h时CarE活力最大(36.75U/mg);甲维盐对CarE活力表现先抑制后诱导的作用,在72h时CarE活力最大(42.44U/mg)。2种农药对GST活性在72h内的变化一致,均呈现“诱导-抑制-诱导”的规律。氯虫苯甲酰胺处理后CYP450活性与对照相比无显著差异;甲维盐处理36h、48h、72h幼虫体内CYP450活性均显著低于对照。
To clarify the toxicity and detoxification mechanism of chlorantraniliprole and emamectin benzoate to Spodoptera frugiperda ,the LC 10 ,LC 20 and LC 50 of two insecticides against third instar larvae were determined by leaf impregnation method,and the effects on the activities of the three main detoxifying enzymes carboxylesterase (CarE),glutathione-S-transferase (GST),cytochrome P450 (CYP450) of larvae were explored by two pesticides with the sublethal dose (LC 20 ).The results showed that the toxicity of methyvitamin was higher to larvae,and the values of LC 10 ,LC 20 and LC 50 were 0.005,0.007 and 0.013 mg/L at 48 h,respectively.During 72 hours of treatment with two pesticides,the activity of three enzymes of larvae showed different dynamic changes.CarE activity was first induced and then inhibited by chlorantraniliprole,and CarE activity was the highest (36.75 U/mg) at 24 h.CarE activity was first inhibited and then induced by emamectin benzoate,and CarE activity was the highest (42.44 U/mg) at 72 h.The changes of GST activity of the two pesticides were consistent within 72 h,showing the rule of “induction-inhibition-induction”.The activity of CYP450 in larvae treated with chlorantraniliprole was not significantly different from that of the control,and the activity of CYP450 in larvae treated with emamectin benzoate for 36,48 and 72 hours was significantly lower than that of the control.
