氟哌利多对氯胺酮诱导的小鼠条件性位置偏爱的影响

摘要 目的观察氟哌利多对氯胺酮诱导的小鼠条件性位置偏爱的影响。方法将48只小鼠分4组(n=12):生理盐水组(NS组)、氯胺酮组(KT组)、5mg/kg氟哌利多+氯胺酮组(KD1组)、10mg/kg氟哌利多+氯胺酮组(KD2组)。接受条件性位置偏爱实验,观察分析受试小鼠用药前后的位置偏爱情况。结果与基础值相比,在戒断24h和48h后,KT组小鼠在白箱停留时间明显延长(P<0.01),KD2组小鼠在白箱停留时间无延长(P>0.05),KD1组小鼠仅在戒断24h后延长(P<0.05);与NS组比较,KT组小鼠在白箱停留时间延长(P<0.05),KD2组小鼠则无差异(P>0.05);与KT组比较,KD2组小鼠在白箱停留时间明显缩短(P<0.01)。结论氯胺酮可诱导小鼠产生条件性位置偏爱,并且一定剂量的氟哌利多能够减弱或取消小鼠对氯胺酮的精神依赖性。 Objective To observe the influence of droperidol at different doses on the Conditional Place Preference ( CPP) induced by ketamine in mice.Methods Forty - eight Kunming mice were randomly divided into four groups ( n = 12): normal saline group ( Group NS),ketamine group ( Group KT),5mg /kg droperidol + ketamine group ( Group KD1),10 mg /kg droperidol + ketamine group ( Group KD2).The residence time in white box were recorded and compared by CPP test.Results Compared with the basic average value within group,the residence time of Group KT prolonged significantly ( P < 0.01) and of Group KD2 had no statistical difference ( P > 0.05) after 24 hours and 48 hours of withdrawal,but that of Group KD1 prolonged ( P < 0.05) only after 24 hours of withdrawal;Compared with Group NS,the residence time of Group KT prolonged ( P < 0.05) but of Group KD2 had no statistical difference ( P > 0.05);Compared with Group KT,the residence time of Group KD2 shortened significantly ( P < 0.01).Conclusions Ketamine can induce CPP in mice and a certain dose of droperidol can reduce or cancel the mental dependence of mice induced by ketamine.
