摘要
目的:研究骨碎补提取物治疗去卵巢骨质疏松大鼠的效果,并观察大鼠血清学指标的变化。方法:将75只雌性SD大鼠随机分为假手术组,模型组,西药组,骨碎补低、高剂量组5组,每组15只。建立去卵巢骨质疏松大鼠模型,假手术组不切除卵巢。在手术后第5周开始,西药组给予戊酸雌二醇灌胃,剂量为0.1 mg/(kg·d);骨碎补低、高剂量组给予骨碎补提取物灌胃,剂量分别为10.0 mg/(kg·d)、40.0 mg/(kg·d);假手术组和模型组大鼠给予等容积的生理盐水,连续给药12周。比较各组大鼠股骨和腰椎骨密度(Bone mineral density,BMD)、血清骨钙素(Osteocalcin,OC)、骨碱性磷酸酶(Bone alkaline phosphatase,BALP)、骨保护素(Osteoprotegerin,OPG)、内皮素-1(Endothelin-1,ET-1)和血管内皮生长因子(Vascular endothelial growth factor,VEGF)水平。结果:与假手术组比较,模型组大鼠股骨和腰椎BMD明显降低,血清OC、BALP、ET-1水平明显升高,OPG、VEGF水平明显降低,差异均有统计学意义(P<0.05)。与模型组比较,西药组及骨碎补低、高剂量组大鼠股骨和腰椎BMD、血清OPG和VEGF水平均升高,血清OC、BALP、ET-1水平明显降低,差异均有统计学意义(P<0.05);与西药组比较,骨碎补高剂量组大鼠股骨和腰椎BMD、血清OPG和VEGF水平较高,血清OC、BALP、ET-1水平较低,差异均有统计学意义(P<0.05)。结论:高剂量骨碎补提取物治疗去卵巢所致骨质疏松大鼠,能够通过调节ET-1、VEGF水平,改善骨代谢,提高BMD。
Objective:To study the effect of extract of Rhizoma Drynariae on ovariectomy-induced osteoporotic rats, and to observe the changes of serological indexes in rats. Methods : A total of 75 female SD rats were randomly divided into five groups, namely the sham operation group, the model group, the western medicine group and the Rhizoma Drynariae groups of low dose and high dose, 15 rats in each group. The models of ovariectomy-induced osteoporotic rats were established, and the ovaries in the sham operation group were not removed. From the 5 th week after operation, the western medicine group was given 0. 1 mg/( kg·d) of estradiol valerate by gavage;the Rhizoma Drynariae groups of low dose and high dose were given 10. 0 mg/( kg·d) and 40. 0 mg/( kg·d) of extract of Rhizoma Drynariae by gavage respectively;the sham operation group and the model group were given saline of the same volume;all groups were administrated for 12 weeks. The levels of bone mineral density( BMD) of femur and lumbar vertebrae, osteocalcin( OC) in serum, bone alkaline phosphatase( BALP), osteoprotegerin( OPG), endothelin-1( ET-1) and vascular endothelial growth factor( VEGF) in rats in each group were compared. Results : Compared with those in the sham operation group, the BMD of femur and lumbar vertebrae as well as the levels of OPG and VEGF in the model group were significantly decreased, while the levels of OC,BALP and ET-1 in serum in the model group were significantly increased, differences being significant( P < 0. 05).Compared with those in the model group, in the western medicine group and the Rhizoma Drynariae groups of low dose and high dose, the BMD of femur and lumbar vertebrae as well as the levels of OPG and VEGF in serum were increased,while the levels of OC, BALP and ET-1 in serum were significantly decreased, differences being significant( P < 0. 05);compared with those in the western medicine group, in t he Rhizoma Drynariae group of high dose,the BMD of femur and lumbar vertebrae as well as the levels of OPG and VEGF in s
