摘要
为建立羊乳中牛乳成分的检测方法,采用西北农林科技大学动物医学院免疫学实验室制备并保存的牛α酪蛋白单克隆抗体,分别建立间接ELISA和间接阻断ELISA检测方法,并对两种方法的灵敏度及重复性进行试验。结果显示,建立的间接ELISA方法测定羊乳中牛α酪蛋白的检测灵敏度为1.3μg/mL,变异系数为2.7%;建立的间接阻断ELISA方法测定羊乳中牛α酪蛋白标准曲线的灵敏度为13.7 ng/mL,变异系数为2.9%。表明间接阻断ELISA标准曲线具有更高的检测灵敏度,对羊乳中牛乳成分的检测更准确。
In order to detect bovine milk components in goat milk,mAb of anti-bovine α-casein prepared by Immunology Laboratory of Northwest A&F University was used to establish an indirect ELISA and an indirect-blocking ELISA method.The results showed that,the sensitivity of indirect ELISA for determining bovine α casein content in goat milk is 1.3μg/mL,and the coefficient of variation(CV)is 2.7%.The sensitivity of indirect-blocking ELISA for determining bovine α casein content in goat milk is 13.7 ng/mL,and the CV is 2.9%.The indirect-blocking ELISA has higher detection sensitivity,and it is more accurate to determine bovine milk components in goat milk.
