摘要
探索大孔树脂对信阳绿茶中多糖的最佳纯化条件。通过对八种不同型号大孔树脂的吸附量、吸附率及解析率进行考察,优选最佳纯化树脂。AB-8型为最优树脂,最佳上样条件为:pH=1.0、上样浓度为5mg/mL、上样速度为2.0BV/h;最佳洗脱条件为:70%的乙醇洗脱液、洗脱剂用量为3.0BV、流速为1.0BV/h。经过该工艺纯化后,绿茶中多糖的纯度由10.6%提高到了64.3%。得出AB-8型大孔树脂能够很好的富集、纯化绿茶中的多糖,为更高效的利用绿茶资源提供了理论依据。
To explore the best purification conditions of polysaccharide in Xinyang green tea by macroporous resin.In this paper,the adsorption capacity,adsorption rate and analytical rate of eight kinds of macroporous resins were investigated,and the best purified resin was selected.The macroporous resin AB-8 is the best,the best loading conditions are:pH=1.0,loading concentration of 5mg/mL,loading speed of 2.0bv/h;the optimal elution conditions were 70%ethanol eluent,3.0bv eluent dosage,1.0bv/h flow rate.After the process,the purity of polysaccharide in green tea increased from 10.6%to 64.3%.The results showed that AB-8 macroporous resin can enrich and purify polysaccharides in green tea,which provides a theoretical basis for more efficiency utilization of green tea resources.
