摘要
【目的】探究蓝果忍冬主栽品种‘L4’、‘Berel’在果实采摘后不同发育时期叶片中的类黄酮种类、含量及抗氧化活性的变化,比较两品种之间的差异,确定蓝果忍冬叶片的最佳采收时期,为蓝果忍冬叶片的合理开发利用提供科研依据。【方法】以果实采摘后不同发育时期(06-09-08-06)的‘L4’、‘Berel’叶片为试料,采用液相色谱与质谱联用法对不同发育时期2个品种叶片的类黄酮种类及含量进行测定,并测定叶片对DPPH·自由基的清除能力和对铁离子的还原能力(FRAP)。【结果】(1)从蓝果忍冬品种‘L4’叶片中分离鉴定出了11种类黄酮成分;从‘Berel’叶片中分离鉴定出10种类黄酮成分,与‘L4’相比缺少了山奈酚3-O-芸香糖苷。槲皮素苷为‘L4’和‘Berel’叶片中主要的类黄酮化合物,占总类黄酮含量的56%~57%。(2)06-23采摘的‘L4’、‘Berel’叶片中的总类黄酮含量最高,分别为37.21和20.61mg/g,显著高于其他时期;同时该时期叶片的抗氧化活性均显著高于其他时期,‘L4’、‘Berel’对DPPH·自由基的清除能力分别为51.90,25.07mg/g,FRAP分别为125.66,91.73mg/g。(3)叶片提取物的抗氧化活性与叶片中的总类黄酮含量呈显著正相关。【结论】‘L4’叶片中的类黄酮种类、含量及抗氧化能力均优于‘Berel’,可作为一种天然类黄酮提取的良好来源,06-23是蓝果忍冬叶片的最佳采收期。
【Objective】This study investigated the types,contents and antioxidant activities of flavonoids in leaves of blue honeysuckle(Lonicera caerulea L.)‘L4’and‘Berel’at different developmental stages after fruit picking and compared differences in two varieties to provide basis for determining optimal harvesting period of blue honeysuckle leaves and its rational development and utilization.【Method】The leaves of‘L4’and‘Berel’at different developmental stages after fruit picking(June 09-August 06)were selected,and the liquid chromatography and mass spectrometry were used to determine types and contents of flavonoids of different varieties at different developmental stages.The DPPH free radical scavenging ability and iron ion reducing ability(FRAP)of the leaves were also determined.【Result】(1)A total of 11 flavonoids were isolated and identified in‘L4’leaves and 10 were identified in‘Berel’leaves,the latter lacked kaempferol 3-O-rutinoside.Quercetin was the main flavonoid compound in both varieties,accounting for 56%-57%of total flavonoids.(2)The total contents of flavonoids in leaves of‘L4’and‘Berel’picked on June 23 were 37.21 and 20.61 mg/g,significantly higher than other periods.The antioxidant activity in leaves in June 23 was significantly higher than that in other periods.The scavenging abilities of‘L4’and‘Berel’against DPPH·free radicals were 51.90 and 25.07 mg/g,and FRAP values were 125.66 and 91.73 mg/g,respectively.(3)Antioxidant activity of leaf extract was significantly positively correlated with total flavonoid content(TFC)in leaves.【Conclusion】The types,contents and antioxidant capacities of flavonoids in‘L4’leaves were better than in‘Berel’leaves,and can be used as a good source of natural flavonoid extraction.June 23 was the best harvesting period for blue honeysuckle leaves.
