补气助阳汤联合乳酸杆菌对变应性鼻炎大鼠鼻黏膜炎症反应及TH1/TH2细胞因子网络表达的影响 被引量:3

摘要 目的:分析补气助阳汤联合乳酸杆菌对变应性鼻炎(AR)大鼠鼻黏膜炎症反应及TH1/TH2细胞因子网络表达的影响。方法:选取SPF级Wistar雄性大鼠40只,随机数字表法将大鼠分成四组,正常组、模型组、乳酸杆菌组及联合用药组,每组各10只。除正常组外,其他各组大鼠制备AR模型,第15天起乳酸杆菌组大鼠灌胃2ml乳酸杆菌稀释液,联合用药组在乳酸杆菌组基础上灌胃2ml补气助阳汤药液,正常组和模型组大鼠灌胃等剂量生理盐水,每天1次,共灌胃2周;观察造模和给药期间各组大鼠饮食、活动及体征变化状况、肠黏膜组织的病理状况,ELISA法检测血清γ-干扰素(INF-γ)、白细胞介素-4(IL-4)、IL-6含量。结果:和正常组对比,模型组大鼠搔抓鼻部与打喷嚏次数上升;和模型组相比,乳酸杆菌组与联合用药组大鼠搔抓鼻部与打喷嚏次数下降,差异均有统计学意义(P<0.05);和正常组相比,模型组大鼠血清IL-4、IL-6含量上升,IFN-γ含量下降;和模型组相比,乳酸杆菌组和联合用药组大鼠血清IL-4、IL-6含量下降,IFN-γ含量上升;和乳酸杆菌组相比,联合用药组大鼠血清IL-4、IL-6含量下降,IFN-γ含量上升,差异均有统计学意义(P<0.05)。结论:补气助阳汤联合乳酸杆菌可降低AR大鼠鼻黏膜内炎症反应程度,改善失衡的TH1/TH2细胞因子网络。 Objective:To analyze the effect of Buqi Zhuyang Decoction combined with Lactobacillus on nasal mucosal inflammation and TH1/TH2 cytokine network expression for rats with allergic rhinitis(AR).Methods:Forty Wistar male rats of SPF grade were randomly divided into four groups:normal group,model group,lactobacillus group and combination group,with 10 rats in each group.In addition to the normal group,AR rats were prepared in the other groups.On the 15 th day,2 ml of Lactobacillus dilution was administered to the rats in the Lactobacillus group,and 2 ml of Buqi Zhuyang Decoction solution was administered by the combination group on the basis of the Lactobacillus group.Rats in the normal group and the model group were intragastrically administered with normal saline once a day for 2 weeks.The changes of diet,activity and signs,and the pathological condition of intestinal mucosa were observed during the modeling and administration.The levels of serumγ-interferon(INF-γ),interleukin-4(IL-4)and IL-6 were detected by ELISA.Results:Compared with the normal group,the number of sneezing and sneezing in the model group increased.Compared with the model group,the number of sneezing and sneezing in the lactobacillus group and the combination group decreased.The significance of learning(P<0.05).Compared with the normal group,the serum IL-4 and IL-6 levels in the model group increased,and the IFN-γcontent decreased;compared with the model group,the lactobacillus group and the combination group rats serum levels of IL-4 and IL-6 decreased,and IFN-γlevels increased.Compared with the Lactobacillus group,serum IL-4 and IL-6 levels decreased,and IFN-γlevels increased in the combination group,academic significance(P<0.05).Conclusion:Buqi Zhuyang Decoction combined with Lactobacillus can reduce the degree of inflammatory reaction in nasal mucosa of AR rats and improve the imbalance of TH1/TH2 cytokine network.
