摘要
从安徽省及周边的7个规模化绵羊场和10个规模化山羊场分别采集832和781份新鲜粪样,采用基于贾第虫磷酸丙糖异构酶(tpi)基因的巢式PCR扩增,随后对获得的tpi基因阳性的样本进行了谷氨酸脱氢酶(gdh)基因的扩增,并对获得的阳性扩增产物进行测序和生物信息学分析。绵羊贾第虫感染率为0.36%,山羊贾第虫感染率为0.00%。tpi基因序列分析显示3个阳性样品基因型分别为E1(2)和E6(1),gdh基因序列分析显示2个阳性样品均属聚集体E。本研究中,安徽及周边地区绵羊和山羊贾第虫感染率较低,且存在的基因型均为反刍动物常见的聚集体E,未发现具有人兽共患潜力的A或B型。
To investigate the prevalence and assemblages of G.duodenalis in sheep and goats in Anhui province and adjacent provinces.A total of 832fecal samples from seven sheep farms and 781 fecal samples from ten goat farms were collected in this study.G.duodenalis was detected using a nested-PCR targeting tpi gene.The identified tpi-positive fecal samples were subjected to a heminested- PCR targeting gdh gene.The all positive PCR products were sequenced,and aligned to determine the assemblages of G.duodenalis.The prevalence for G.duodenalis in sheep and goats were 0.36%(3/832)and 0.00%(0/781)in the present study,respectively.Sequence analysis of tpi gene showed that the three G.duodenalis isolate belong to assemblages E1(2)and E6(1),sequence analysis of gdh gene indicated that the two G.duodenalis isolate belong to assemblages E. These findings show a relatively low occurrence of G.duodenalis in sheep and goats in Anhui province and adjacent provinces.The genotypes obtained in this study were common in domestic ruminants, and there are no‘zoonotic’assemblages.
