抗风竤·东北雌性红萝卜浓缩超微原粉对AGA大鼠模型抗氧化作用及对Nrf2通路的影响 被引量:5

摘要 目的:通过Nrf2参与的Keap1-Nrf2-ARE信号通路探讨抗风竤·东北雌性红萝卜浓缩超微原粉(以下简称为抗风竤)对尿酸钠致急性痛风性关节炎模型大鼠的抗氧化作用,通过实验研究观察踝关节肿胀度,大鼠血清内抗氧化酶SOD、CAT、GSH-Px活力及滑膜内Nrf2表达的改变。方法:通过大鼠右踝胫跗关节注射MSU溶液,建立AGA大鼠模型,安康信组、抗风竤低剂量组、中剂量组和高剂量组,每天灌胃给药1次,空白组、模型组予等量的生理盐水灌胃,观察造模后4、24、48、72h各组治疗前后关节肿胀程度的变化。检测造模72h后大鼠血清内SOD、CAT、GSH-Px的活性;检测大鼠踝关节滑膜组织中Nrf2表达的平均光密度值。结果:(1)造模后4h抗风竤的低、中剂量组与模型组比较肿胀有所减少,尤其是高剂量组减少明显。(2)与模型组比较,安康信组,抗风竤低、中、高剂量组的SOD活力明显增加;与模型组比较,抗风竤高剂量组CAT活力增加;与模型组比较,安康信组、抗风竤高剂量组GSH-Px活力明显增加,抗风竤中剂量组GSH-Px活力增加。(3)大鼠右踝关节滑膜组织中Nrf2,与模型组相比,安康信组及抗风竤中、高剂量组的大鼠右踝关节滑膜组织中Nrf2的阳性染色区平均光密度值明显增加(P<0.01),低剂量组增加(P<0.05)。结论:(1)抗风竤能够减轻痛风性关节炎大鼠踝关节肿胀度。(2)抗风竤具有提高AGA大鼠模型体内抗氧化酶SOD、CAT、GSH-Px的活力。(3)抗风竤使AGA大鼠模型机体内Nrf2表达水平上调。 Objective:Through the Keap1-Nrf2-ARE signaling pathway involved in Nrf2,the antioxidative effect of Kangfenghong·northeast female red radish concentrate ultrafine original powder(hereinafter referred to as Kangfenghong)on rats model with acute gouty arthritis induced by sodium urate was studied.To observe ankle swelling degree,through experiment in rat serum SOD,CAT and GSH-Px activity of antioxidant enzymes and synovial Nrf2 in expression change.Methods:Through rat right ankle shank hocks MSU injection solution,AGA rat model was established,etoncoxib tablets group,Kangfenghong low dose group,middle dose group and high dose group,irrigation to 1 times a day,the blank group,model group in the same amount of saline lavage,observe the building after 4 h,24 h,48 h,72 h each joint swelling degree of change before and after the treatment.Detection of building 72 h after rat model serum SOD,CAT and GSH-Px activity;Detection of Nrf2 expression in rat ankle synovial tissue,the average optical density value.Results:(1)4 h after building model Kangfenghong low dose group、middle dose group compared with model group,swelling alleviate,especially in high dose group reduced obviously;(2)Compared with the model group,the SOD activity of the etoncoxib tablets group,the Kangfenghong low,medium and high dose groups increased significantly.Compared with the model group,the Kangfenghong high dose group CAT activity increased.Compared with the model group,the GSH-Px activity of the etoncoxib tablets group and the Kangfenghong high dose group increased significantly.The Kangfenghong medium dose group GSHPx vitality increased.(3)Compared with the model group,the average optical density of Nrf2 in the positive stain area of the right ankle synovial tissue of rats in the etoncoxib tablets group,Kangfenghong middle dose group and high-dose group was significantly increased(P<0.01),while that of the low-dose group was increased(P<0.05).Conclusion:(1)Kangfenghong can reduce the ankle swelling of gout arthritis rats.(2)Kangfenghong can imp
