摘要
半胱氨酸蛋白酶-1(cysteine-aspartic proteases-1,caspase-1)是一种半胱氨酸天冬氨酸特异性蛋白水解酶,有活性的半胱氨酸蛋白酶可对天冬氨酸蛋白酶的特异性位点进行切割,使蛋白质裂解。当病原微生物感染或者存在其他危险信号时,有活性的caspase-1可对执行蛋白GSDMD的N端和C端结构域进行切割,活化的N-结构域可促进细胞膜形成亲水性孔道,导致细胞膨胀裂解,从而促进白细胞介素(interleukin,IL)-1β和IL-18的释放引发一系列级联反应,参与机体炎症反应、细胞焦亡的过程。近年研究表明,caspase-1活化是眼科多种疾病发病机制的中心环节,尤其在角膜疾病,年龄相关性白内障,糖尿病视网膜病变,年龄相关性黄斑变性等疾病中作用重大,了解caspase-1在眼科疾病中的作用对相关疾病的诊治很有意义。
Caspase-1 is a cysteine aspartate-specific proteolytic enzyme,As an active cysteine protease,Caspase-1 can cleave a specific site for aspartic protease,resulting in cleaved protein.When pathogenic microorganisms are infected or other dangerous signals are presented,the active caspase-1 can cleave the N-terminal and C-terminal domains of the protein GSDMD,and the activated N-domain can promote the formation of hydrophilic pores in the cell membrane,resulting in cells expansion and cleavage,which promotes the release of(interleukin)IL-1βand IL-18,leads to a series of cascade reactions,which are involved in the process of inflammatory reaction and cell death.Recent studies have shown that caspase-1 is the central link in the pathogenesis of ophthalmology,especially in corneal diseases,age-related cataract,diabetic retinopathy,age-related macular degeneration and other diseases.To know the role of in ophthalmic diseases is great significance for the diagnosis and treatment of related diseases.
