摘要
本文旨在对不同生长期串叶松香草超氧化物歧化酶(SOD)提取条件进行优化,并对其提取液中类SOD活性物质及抗氧化能力进行考察。采用单因素和正交试验方法,以SOD活力为标准,优化提取条件,并测定串叶松香草不同生长阶段总黄酮、多酚类化合物和维生素C等类SOD活性物质含量,同时对不同生长阶段提取液中羟自由基、1,1-二苯代苦味酞基(DPPH)自由基及超氧阴离子的清除能力进行测定。结果表明:叶丛期与开花期的水提条件为料液比1∶25,37℃水浴浸提12h,其提取液SOD活性分别为4030.12、4445.31U/g。与叶丛期相比,开花期维生素C、总黄酮和总多酚含量分别提高34.51%(P<0.01)、56.59%(P<0.01)和23.15%(P<0.01),开花期提取液清除羟自由基和DPPH的能力分别提高205.65%(P<0.01)和12.20%(P<0.01)。综上,串叶松香草开花期比叶丛期抗氧化能力更强。
This study was to investigate best condition for extraction superoxide disinutase(SOD) from different growth phase of Silphium perfoliatum and evaluate the effects of SOD like activity substances and antioxidant activity of extract from Silphium perfoliatum.Use SOD activity as the standard to optimize extraction condition by the single factor experiments and orthogonal experiments.The content of SOD-simultaneous active substances of Silphium perfoliatum were determided. Determination of flavonoids,polyphenols and vitamin C(VC) content by standard curve method.Meanwhile,put the liquid antioxidant capacity to take preliminary determination,including its hydroxyl radical·OH,DPPH and superoxide anion scavenging capacity in different growing stage.The results showed that in the condition that solid-liquid ratio 1: 25,extaction temperature 37 ℃,extraction time 12 h,the SOD in Silphium perfoliatum water extract of the highest activity (4030.12 U/g and 4445.31 U/g) in the leaf stage and flowering stage.Compared with the leaf stage,the contents of VC,total flavonoid and total polyphenols in water extracts of Silphium perfoliatum were significantly increased by 34.51%(P < 0.01),56.59%(P < 0.01) and 23.15%(P < 0.01).Meanwhile, the ability of hydroxyl radicals and DPPH scavenging in water extracts of Silphium perfoliatum at the flowering stage were increased by 205.65%(P < 0.01) and 12.20%(P < 0.01) than the leaf stage.
