摘要
目的研究小鼠失血性休克后脓毒症急性肺损伤CD200分子的表达。方法采用失血性休克后盲肠末端结扎穿孔复制小鼠急性创伤后脓毒症急性肺损伤模型。将20只小鼠随机分为急性肺损伤组和假手术组,于盲肠末端穿孔术后24 h取小鼠心脏血,肺组织。流式细胞术检测小鼠血液中巨噬细胞、中性粒细胞表面CD200表达;ELISA检测肺组织中TNF-MIP-2、IL-10表达水平;测定肺湿/干质量比;RT-PCR检测肺组织CD200 mRNA表达;Western blot检测术后4,12,24,48 h肺组织CD200表达水平。结果急性肺损伤组巨噬细胞和中性粒细胞CD200表达均较假手术显著升高(P<0.01);细胞因子TNF-、MIP-2、IL-10表达均显著升高(P<0.01);急性肺损伤组湿/干质量比显著高于假手术组(P<0.01);急性肺损伤组CD200 mRNA表达显著高于假手术组(P<0.01);术后4 h肺组织CD200蛋白表达水平显著低于假手术组,24 h与48 h均显著高于假手术组(P<0.05)。结论CD200分子参与失血性休克后脓毒症引起急性肺损伤。
Objective To investigate the expression profiles of CD200 in hemorrhage induced priming for acute lung injury subsequent to septic challenge.Methods Twenty C57/BL mice were divided into sham-hemorrhage(Hem)/cecal ligation and puncture(CLP) and Hem/CLP groups. Blood was collected via cardiac puncture at 24 h post-CLP, at the same time, lung tissue was harvested. CD200 expression on monocytes and neutrophil in blood were measure by flow cytometry. ELISAs for TNF-(,MIP-2 and IL-10 were performed as per manufacturer’s protocol on lung tissue. Lung tissues were weighted at wet/dry conditions. RT-PCR was applied to detect the CD200 mRNA expression. The protein of CD200 in lung were measured at 4, 12, 24, and 48 h post-CLP.Results The expression profiles of CD200 on monocytes and neutrophil were significantly higher response to Hem/CLP(P<0.01). Mice subjected to Hem/CLP, showed a significant increase in lung tissue TNF-(, MIP-2, IL-10 compared with their respective sham-Hem/CLP. Level of lung tissue wet/dry was assessed higher in Hem/CLP group. In those Hem/CLP mice, there was a significant increase in lung tissue CD200 mRNA levels vs sham-Hem/CLP. The protein level of CD200 was significantly decreased at 4 h and increased at 24 and 48 h post-Hem/CLP.Conclusion CD200 was involved in the progression of shock/sepsis induced acute lung injury.
