摘要
目的:利用Lewis肺癌皮下移植肿瘤模型探讨CD4^+CD25^+Treg细胞在肺癌的生长与转移中的作用。方法:利用C57BL/6鼠建立Lewis肺癌皮下移植肿瘤模型;流式细胞仪技术分析荷瘤鼠脾脏中CD4^+CD25^+Foxp3^+Treg细胞的百分比和数量;过继回输CD4^+CD25^+Treg细胞观察肿瘤的生长,HE组织染色技术判明肺部转移性肿瘤结节;CD4^+CD25^+Treg细胞与LLC细胞共培养方法分析CD4^+CD25^+Treg细胞的作用机制。结果:荷瘤鼠脾脏中CD4^+CD25^+Foxp3^+Treg细胞的细胞比例和数量均明显增加。与对照组荷瘤鼠相比,过继回输CD4^+CD25^+Treg细胞后荷瘤鼠的肿瘤体积相对增大,生存缩短,生存率显著降低;HE组织染色技术显示过继回输CD4^+CD25^+Treg细胞后荷瘤鼠的肺部转移性肿瘤结节数量显著增多。细胞共培养及划痕实验结果显示,CD4^+CD25^+Treg细胞非直接促进LLC细胞的迁移。结论:CD4^+CD25^+Treg细胞以非直接作用于肿瘤细胞方式促进Lewis肺癌的肿瘤生长与转移。
Objective: To investigate the role of CD4^+CD25^+Treg cells in the growth and metastasis of lung cancer by using Lewis lung cancer subcutaneous transplantation tumor model. Methods: The transplantation tumor model of Lewis lung cancer was established by subcutaneous injection of LLC cells to C57BL/6 adult mice. The percentage and quantity of CD4^+CD25^+Foxp3^+Treg cells in the splenocytes of tumor-bearing mice were analyzed by flow cytometry. Adoptive transfer of The tumors tumor were observed after adoptive transfer of CD4^+CD25^+Treg cells,HE tissue staining was to identify pulmonary metastatic tumor nodules;CD4^+CD25^+Treg cells and LLC cells co-culture was established to analyze the mechanism of function of CD4^+CD25^+Treg cells. Results: The frequency and absolute number of CD4^+CD25^+Foxp3^+Treg cells increased significantly in the splenocytes of tumor-bearing mice. Compared with the control mice,the tumor volume of tumor-bearing mice was greatly increased after adoptive transfer of CD4^+CD25^+Treg cells,and the survival period of tumor-bearing mice was shortened,and the survival rate was significantly decreased. HE tissue staining revealed that adoptive transfer of CD4^+CD25^+Treg significantly increased the number of lung metastatic tumor nodules in the tumor-bearing mice. Cell co-culture and scratch test showed that CD4^+CD25^+Treg cells did not directly promote the migration of LLC cells. Conclusion: CD4^+CD25^+Treg cells promote tumor growth and metastasis of Lewis lung cancer independent on direct interaction with tumor cells.
