摘要
目的观察冠心平对动脉粥样硬化(AS)模型ApoE^-/-小鼠血清氧化型低密度脂蛋白(ox-LDL)、氧化型低密度脂蛋白受体(LoX)-1mRNA、细胞间黏附分子(ICAM)-1、内皮细胞白细胞黏附分子(ELAM)-1水平的影响。方法将AS模型ApoE^-/-小鼠分为模型组(蒸馏水灌胃)、阿托伐他汀(6.67mg/kg)组、苯磺酸氨氯地平(1.67mg/kg)组、冠心平低剂量(0.75g/kg)组、冠心平高剂量(1.50g/kg)组,同时设正常组(喂养基础饲料)。喂养3个月后,采用酶联免疫吸附实验(ELISA)检测各组ox-LDL的浓度,Western印迹法测定ELAM-1、ICAM-1及LoX-1蛋白表达水平,用聚合酶链式反应(PCR)检测LoX-1mRNA水平。结果高剂量冠心平可明显降低AS模型ApoE^-/-小鼠ox-LDL、LoX-1、LoX-1mRNA、ICAM-1及ELAM-1水平,且高剂量冠心平抑制LoX-1作用优于阿托伐他汀、氨氯地平;低剂量冠心平对LoX-1也有明显的抑制作用。结论冠心平可有效降低AS模型ApoE^-/-小鼠ox-LDL、LoX-1、LoX-1mRNA、ICAM-1、ELAM-1水平,具有抗AS作用。
Objective To observe the effect of Guanxinping on the levels of ox-LDL,LoX-1 protein and mRNA,ICAM-1,ELAM-1 of ApoE^-/-mice with atherosclerosis.Methods ApoE^-/-model mice were randomly divided into model,atorvastatin〔6.67 mg/(kg·d)〕,amlodipinebesylate〔1.67mg/(kg·d)〕,Guanxinping low dose〔0.75 g/(kg·d)〕,high dose〔1.5 mg/(kg·d)〕and normal groups.After 3 months,the levels of serum ox-LDL,LoX-1 protein and mRNA,ICAM-1,ELAM-1 were measured.Results High dose Guanxinping decreased the levels of ox-LDL,LoX-1 protein and mRNA,ICAM-1,ELAM-1 of ApoE^-/-mice with atherosclerosis,and the effects of high-dose Guanxinping inhibition of LoX-1 were better than those of atorvastatin and amlodipinebesylate,and low dose Guanxinping inhibited LoX 1 significantly.Conclusions Guanxinping could effectively reduce the levels of serum ox-LDL,LoX-1 protein and mRNA,ICAM-1,ELAM-1 of ApoE^-/-mice with atherosclerosis,and has anti-atherosclerosis effect.
