Optimization of Flash Extraction of Total Polyphenols from Leaves of Eriobotrya japonica Lind by Response Surface Methodology

摘要 以总多酚得率为指标,在单因素实验基础上,采用Box-Behnken响应面法对影响枇杷叶总多酚提取得率的乙醇质量分数、提取时间、料液比等主要因素进行优化。优化的枇杷叶总多酚闪式提取工艺条件为:固定提取次数2次,采用32倍量质量分数62%乙醇溶液作为提取溶剂,闪式提取127s,在此工艺条件下,枇杷叶总多酚得率为(33.22±1.6)mg·g-1,与模型预测值32.86mg·g-1相近,并且远高于回流提取及超声提取条件下总多酚得率。响应面法优选的枇杷叶总多酚闪式提取工艺稳定,提取时间短、效率高,适合于工业化生产,实验结果对枇杷叶的规模化开发利用具有一定的参考价值。 With the extraction rate of total polyphenols as response value, the ethanol concentration, extraction time, liquid to material ratio were optimized by Box-Behnken response surface methodology (RSM ) based on the results of single-factor experiment. The optimal conditions obtained by RSM were as follows: fixed extraction times was 2 times,ethanol concentration was 62%, liquid to material ratio was 32, extraction time was 127 s. Under these conditions, the experimental yield of total polyphenols was (33.22±1.6 ) mg·g -1 , which was well matched with the predictive yield of 32.86 mg·g -1 . Furthermore, flash extraction presented highest extraction rate compared with reflux extraction and ultrasonic extraction. The result revealed that flash extraction was simple, rapid and efficient, and the optimized extraction technology could provide the scientific basis for industrialized development.
