Beclin 1在乳腺浸润性导管癌中的表达及其与Ki67的关系 被引量:3

摘要 目的探讨自噬相关基因Beclin 1和增殖相关基因Ki67编码的蛋白在乳腺浸润性导管癌中的表达情况。方法选取乳腺浸润性导管癌患者64例。采用免疫组化法检测癌组织及对应的癌旁组织中Beclin 1和Ki67蛋白的表达,同时采用免疫荧光法和免疫印迹法检测癌组织及对应癌旁组织中Beclin 1蛋白的表达。结果癌组织中Beclin 1的阳性表达率(43.8%)明显低于癌旁组织(98.5%)(P<0.05),而Ki67的阳性表达率(76.6%)则高于癌旁组织(0.00%)(P<0.05)。癌组织中Beclin 1蛋白表达与Ki67蛋白表达呈负相关(r=-0.256,P<0.05)。Beclin 1蛋白表达与乳腺浸润性导管癌病理分级、有无淋巴结转移有关(P<0.05),与肿瘤直径、年龄、雌激素受体(ER)、孕激素受体(PR)、人类表皮生长因子受体2(HER-2)均无关(P>0.05)。Ki67蛋白表达与乳腺浸润性导管癌病理分级、肿瘤直径有关(P<0.05),与有无淋巴结转移、年龄、ER、PR、HER-2均无关(P>0.05)。免疫荧光法检测结果显示癌组织中Beclin 1蛋白的阳性表达率(30.0%)明显低于癌旁组织(100.0%)(P<0.05)。免疫印迹法检测结果显示癌旁组织中Beclin l蛋白的相对表达量为0.43±0.12,明显高于癌组织(0.20±0.18)(P<0.05)。结论乳腺浸润性导管癌组织中Beclin 1蛋白表达明显降低,Ki67蛋白表达明显升高。 Objective To investigate the expression of autophagy-associated gene Beclin 1 and proliferationrelated gene Ki67 in breast invasive ductal carcinoma.Methods The expressions of Beclin 1 and Ki67 were determined by immunohistochemistry in 64 patients with breast invasive ductal carcinoma and corresponding adjacent tissues.Immunofluorescence and western blotting were used to determine the expressions of Beclin 1 in breast invasive ductal carcinoma and corresponding adjacent tissues.Results The positive expression rate of Beclin 1 in carcinoma tissues(43.8%)was lower than that in adjacent tissues(98.5%)(P<0.05).The positive expression rate of Ki67(76.6%)was higher than that in adjacent tissues(0.00%)(P<0.05).The positive rates of Beclin 1 and Ki67 in carcinoma tissues were negatively correlated(r=-0.256,P<0.05).The expression of Beclin 1 was associated with pathological grade and lymph node metastasis(P<0.05),but it was not associated with tumor diameter,age,estrogen receptor(ER),progesterone receptor(PR)and human epidermal growth factor receptor 2(HER-2)(P>0.05).The expression of Ki67 was associated with pathological grade and tumor diameter(P<0.05),but it was not associated with lymph node metastasis,age,ER,PR and HER-2(P>0.05).The results of immunofluorescence showed that the positive expression rate of Beclin 1 in carcinoma tissues(30.0%)was lower than that in adjacent tissues(100.0%)(P<0.05).The results of western blotting showed that the relative expression of Beclin 1 in adjacent tissues was 0.43±0.12,which was higher than that in carcinoma tissues(0.20±0.18)(P<0.05).Conclusions The expression of Beclin 1 is down-regulated in breast invasive ductal carcinoma,while the expression of Ki67 is up-regulated.
