牙周炎不同阶段Th17/Treg细胞平衡变化初探 被引量:4

摘要 目的 建立实验性大鼠牙周炎模型,观察外周血及牙周组织Th17/Treg细胞平衡在牙周炎不同阶段的变化以及牙槽骨高度变化,为探寻用于研究牙周炎正畸牙齿移动的动物模型提供参考。方法 32只SD雄性大鼠随机分为3个实验组(牙周炎A组、牙周炎B组和牙周炎C组,每组8只)和1个对照组(8只)。实验组采用牙颈部丝线结扎并局部注射LPS的方法建立实验性牙周炎模型后,牙周炎A组处死,采用流式细胞仪检测外周血Th17细胞、Treg细胞比例变化。免疫组化检测牙周组织ROR-γt、Foxp3表达。实时定量PCR检测牙龈组织ROR-γt mRNA、Foxp3 mRNA的表达。牙周炎B组和C组去除局部刺激并行牙周洁治,分别于术后第7、14天进行牙周检查,并行上述流式细胞等检测。结果 牙周炎A、B及C组外周血Th17细胞、Treg细胞、Th17/Treg细胞比例及牙龈组织ROR-γt mRNA、Foxp3 mRNA的表达均较对照组明显升高( P <0.05)。牙周炎B组和C组Th17细胞、Th17/Treg细胞比例及牙龈组织ROR-γt mRNA的表达较牙周炎A组明显降低( P <0.05),但Treg细胞及Foxp3 mRNA表达较牙周炎A组明显升高( P <0.05)。牙周炎B组和C组相比,Th17细胞、Treg细胞及Th17/Treg细胞比例差异无统计学意义( P >0.05)。牙周炎A组、B组、C组牙槽嵴顶距釉牙骨质界距离均较对照组明显增加( P <0.05),但三组间差异无统计学意义( P >0.05)。结论 Th17、Treg细胞及Th17/Treg细胞平衡参与不同阶段牙周炎的免疫调控。局部炎症因素去除后,大鼠牙周炎病变稳定,可以尝试作为研究牙周炎正畸牙齿移动的动物模型。 Objective To investigate the changes of Th17/Treg cell balance in peripheral blood and gingival tissue of rats with periodontitis. Methods Thirty-two male Sprague- Dawley rats were randomly divided into three experimental groups (periodontitis group A, B and C) and one control group, eight rats in each group. Rats in group A, B and C were treated with ligation and local injection of LPS to establish periodontitis model. Then rats in group A were sacrificed and Th17 and Treg cells in peripheral blood were detected by flow cytometry. The changes of ROR-γt and Foxp3 in periodontal tissue were measured using immumohistochemical staining. Realtime PCR was used to measure the expression of ROR-γt mRNA and Foxp3 mRNA in the gingival tissues. The stimulating factors of periodontitis were removed in group B and C, and after 7 and 14 days, the rats in group B and C were sacrificed and same detections performed as those in group A. Results The Th17 cell, Treg cell and Th17/Treg in peripheral blood and the expression of ROR-γt and Foxp3 in periodontal tissues were increased significantly in experimental groups than those in control group ( P <0.05). The Th17 cell and the ratio of Th17/Treg in peripheral blood and the expression of ROR-γt in periodontal tissues in group B and C were decreased compared with group A. The expression of Treg cell and Foxp3 were increased compared with group A. The percentage of Th17, Treg and Th17/Treg showed no difference between group B and group C ( P >0.05). Compared with control group, a significant increase was noted in the distance between cemento- enamal junction and alveolar ridge crest in group A, B and C ( P <0.05). However, there was no significant difference between these three groups. Conclusion The Th17, Treg cell and Th17/Treg were involved in the immunoregulation in different stages of periodontitis. The rats after removing the stimulating factors could be used as the model for studying orthodontic tooth movement with periodontitis.
