摘要
建立C18柱串联CR-I(+)手性冠醚柱的反相高效液相色谱分离方法,用于同时拆分三种芳香氨基酸对映体.研究了流动相比例、流速、柱温以及色谱柱的串联次序对三种芳香氨基酸对映体拆分的影响.实验结果表明:当流动相为pH2.0的高氯酸溶液-乙腈(85∶15,体积比),流速为0.4mL/min,柱温为21℃,串联顺序为手性冠醚柱在前、C18柱在后时三种芳香氨基酸对映体分离效果最优.在最优色谱条件下,分离并测定了米酒和啤酒样品中的三类芳香氨基酸对映体.
A reversed phase chromatographic method of C18 column tandem chiral crown ether column was developed to simultaneously separate three aromatic amino acids enantiomers. The conditions, including mobile phase, flow rate, column temperature and column tandem order, were optimized. The best separation results could be obtained under the conditions of perchloric acid solution(pH 2.0)-acetonitrile(85∶15, volume ratio) mobile phase, 0.4 mL/min flow rate, 21 ℃ column temperature, and the column order of chiral crown ether column tandem C18 column. Three aromatic amino acids in rice wine and beer samples were separated and determined by the proposed method.
