摘要
目的探讨长链非编码RNA-HOTAIR对胃癌细胞增殖凋亡的影响及可能机制。方法将胃癌SGC7901细胞株分为:空白对照组(Blank组)、HOTAIR过表达组(转染HOTAIR过表达质粒)和si-HOTAIR组(转染si-HOTAIR质粒),其中空白对照组为单纯的SGC7901细胞株。qRT-PCR检测细胞中HOTAIR表达量,CCK8、平板克隆形成实验检测细胞增殖情况,流式细胞术及Caspase3/7活性试验检测细胞凋亡情况,qRT-PCR检测各组细胞中微小RNA 126(miR-126)的表达量。结果(1)HOTAIR过表达组细胞增殖水平显著增高,而凋亡率下降;而si-HOTAIR组细胞的增殖水平显著降低,凋亡率上升(P <0.05);(2)胃癌细胞中HOTAIR与miR-126表达量呈负相关(P <0.05)。结论 HOTAIR可促进胃癌SGC-7901细胞的增殖并抑制其凋亡,其机制可能与miR-126表达量有关。
Objective To investigate the effect and possible mechanism of LncRNA-HOTAIR on proliferation and apoptosis of gastric cancer 7901 cells. Methods The human gastric cancer cells(SGC-7901)were divided into blank control group(Blank group),HOTAIR overexpression group and HOTAIR inhibition group(si-HOTAIR group). qRT-PCR was used to detect the relative expression ofHOTAIR in each group. CCK8 and colony formation assay were used to detect the proliferation of cells,and the apoptosis of cells was detected by flow cytometry and Caspase 3/7 activity assay. qRT-PCR was used to detect the relative expression of miR-126 in each group.Results The HOTAIR overexpression group cells showed a significant increase in proliferation and a decrease in apoptotic rate. The proliferation in the si-HOTAIR group cells was significantly inhibited while the apoptosis was increased(P < 0.05). There was a negative correlation between HOTAIR and miR-126 expression in gastric cancer cells(P < 0.05). Conclusion HOTAIR can increase the proliferation and inhibit the apoptotic of gastric cancer7901 cells,and the mechanism may be related to the expression of miR-126.
