Analysis of the expression and the potential of clinical prognostic value of WNT3A,WNT4,WNT11,C-MYC and SFRP2 in ovarian cancer

摘要 目的探讨Wnt信号通路关键因子WNT3A,WNT4,WNT11,C-MYC和SFRP2在卵巢癌中的表达以及其对卵巢癌临床预后的意义。方法采用免疫组织化学方法(IHC)检测不同卵巢癌患者样本中WNTS信号通路关键因子的表达。采用GEPIA分析这些关键因子与卵巢癌的临床分期和预后之间的关系。结果 IHC结果显示WNT3A,WNT4,SFRP2和C-MYC在卵巢癌患者中异常表达(P<0.05)。特别值得关注的是,WNT3A的下游蛋白之一的C-MYC表达上调,而WNT通路的抑制剂SFRP2表达下调。然而,预后分析结果显示,C-MYC,WNT3A,WNT4和SFRP2的表达水平均与卵巢癌临床分期之间无相关性(P>0.05)。但作为预后指标的C-MYC与生存时间显著相关(P<0.05)。C-MYC高表达患者接受治疗比C-MYC表达低的患者的生存时间更长。结论C-MYC是Wnt信号通路的关键因子,并与卵巢癌预后生存时间相关,结果提示其对卵巢癌的预后评价存在价值。本研究为WNTs在卵巢癌中被激活的假设提供了证据。 Objective To investigate the expression of WNT3A, WNT4, WNT11, C-MYC and SFRP2 in ovarian cancer and their clinical significance for ovarian cancer.Methods The expression of key factors of WNT signaling pathway was determined by immunohistochemical (IHC) in different ovarian cancer patient's samples. Subsequently, the relationship between these key factors and the clinical stage and prognosis of ovarian cancer was analyzed using GEPIA.Results IHC results showed that WNT3A, WNT4, SFRP2 and C-MYC were differentially expressed in ovarian cancer patients ( P 〈0.05). Specially, C-MYC was also up-regulated while SFRP2 was down-regulated. However, there has no statistical association between C-MYC, WNT3A, WNT4 and SFRP2 and clinical stage of ovarian cancer based on the correlation analysis generated by GEPIA ( P 〉0.05). As a prognostic marker, survival analysis indicated that only C-MYC shown significant relationship with survival outcome ( P 〈0.05).Conclusion The study provided evidences to show the WNTs were activated in ovarian cancer. C-MYC is a key factor of WNT signaling pathway and is associated with the prognosis of ovarian cancer, indicating that it has value in the evaluation of prognosis of ovarian cancer.
