Cloning and Sequence Analysis of SVP Homologous Gene from Osmanthus Fragrans

摘要 试验以桂花‘日香桂'Osmanthus fragrans cv.rixianggui嫩叶为试验材料,根据转录组数据库中SVP基因的保守序列信息,通过PCR方法克隆得到日香桂SVP基因,将其命名为QfSVP,运用在线生物信息学分析工具对OfSVP序列进行分析。结果表明:OfSVP基因全长1325bp,开放阅读框(ORF)为687bp,编码228个氨基酸。氨基酸序列保守性分析发现,OfSVP所编码的蛋白中含有MADS-box基因家族特有的K-box区域。通过ProtParam ExPASy在线软件分析OfSVP蛋白质分子式为C1121H1859N327O364S9,分子量为26030.60Da,理论等电点PI值为5.80。不稳定指数为68.10,属于不稳定蛋白质。蛋白氨基酸序列比对和系统发育进化分析结果表明OfSVP与油橄榄(Olea europaea)、黄连(Coptis chinensis)和芝麻(Sesamum indicum)的同源性分别达到94%、83%和82%;OfSVP基因的克隆与序列分析对于进一步了解桂花'日香桂'成花机理具有重要意义。 In this study, the young leaves of Osmanthus fragrans cv. rixianggui were used as experimental materials. According to the conserved sequence information of SVP gene in transcriptome database, the SVP gene was cloned by PCR and named as OfSVP. The OfSVP sequence was analyzed by online bioinformatics analysis tool. The results showed that the OfSVP gene was 1325 bp and the open reading frame (ORF) was 687 bp, encoding 228 amino acids. Conservative analysis of amino acid sequences revealed that the protein encoded by OfSVP contains the unique K -box region, which is belonged to the MADS -box gene family. The molecular formula of OfSVP protein was C1121H1859N327O364S9, its relatively molecular weight was 26030.60 kDa and theoretical isoelectric point (PI) was 5.80. The instability index is 68.10, which means it is a unstable protein. Protein amino acid sequence alignment and phylogenetic analysis showed that the homology of OfSVP with Olea europaea, Coptis chinensis and Sesamum indicum reached 94%, 83%, and 82%, respectively. The cloning and sequence analysis of OfSVP gene is of great significance for further understanding of the flowering mechanism of Osmanthus fragrans.
