Preparation of asiaticoside nanoemulsions and quality evaluation

摘要 目的:制备积雪草苷纳米乳(asiaticoside nanoemulsions,ASI-NEs),优化纳米乳处方,并对其理化性质进行考察。方法:本研究以纳米乳的包封率和溶解度作为评价指标,利用单纯形网格法优化处方,并考察最优处方纳米乳的pH、粒径分布、稳定性等理化性质。结果:ASI-NEs的最优处方组成为:单亚油酸甘油酯、RH 40、二乙二醇单乙醚和水,各部分比例依次为5%,15%,26.3%和53.7%。溶解度和包封率实测值与预测值偏差依次为4.55%和3.69%。最优处方的溶解度为13.97 mg·g^(-1),包封率为90.29%。ASI-NEs为O/W型纳米乳,外观澄清透明,pH值为6.8,平均粒径为(132±5.84)nm,多分散指数(PDI)为0.221 1,具有较好的稳定性,高速离心不分层。结论:本研究成功制备了ASI-NEs,且具有良好的理化性质,为纳米乳载体的进一步应用奠定了基础。 Objective:To prepare the carrier of asiaticoside nanoemulsions(ASI-NEs),optimize the prescription,and evaluate the quality.Methods:The simplex lattice design(SLD)was utilized to screen and optimize the formulation,and the drug solubility and encapsulation efficiency were inspected as evaluation indices.The physicochemical properties such as pH value,particle size distribution and stability of the optimal prescription of ASI-NEs were investigated.Results:Following the optimization,the optimal ASI-NEs formulation was composed of 5%glycerol monooleate,15% Cremophor RH 40,26.3% diethylene glycol monoethyl ether and 53.7%water.The deviations between the measured value and the predicted value of solubility and entrapment efficiency were4.55% and 3.69%,respectively.The solubility and encapsulation efficiency of the optimal prescription were 13.97 mg·g^-1 and 90.29%,respectively.ASI-NEs were O/W nanoemulsion with clear and transparent appearance,pH value of 6.8,average particle size of(132±5.84)nm and PDI of 0.221 1.ASI-NEs had good stability,without delamination under the high-speed centrifugation.Conclusion:In this study,ASI-NEs were successfully prepared.ASI-NEs have good physicochemical properties.This study provided theoretical basis for further study of the carrier of nanoemulsions.
