摘要
目的探讨miR-802和RAB23在食管鳞癌中的表达水平及其临床意义。方法选取2014年3月~2015年2月于笔者医院进行食管癌根治手术的标本120例为食管鳞癌组,另选取癌旁正常组织为对照组(120例)。采用实时荧光定量PCR法检测食管鳞状癌组织及癌旁组织中miR-802与RAB23mRNA相对表达量,免疫组化法检测食管鳞状细胞组织中RAB23蛋白表达,根据检测结果将miR-802分为高表达组与低表达组,RAB23分为阳性表达组与阴性表达组,并分析miR-802、RAB23表达与患者临床病理参数的关系,采用Pearson法分析miR-802与RAB23的相关性,Kaplan-Meier法对食管鳞癌患者的生存情况进行分析,并采用Log-Rank法检测不同表达组间生存差异。结果与正常组比较,食管鳞癌组miR-802表达水平显著降低(P<0.05),而RAB23mRNA表达水平显著升高(P<0.05);免疫组化组显示食管鳞癌组RAB23蛋白阳性表达显著高于正常组(P<0.05);临床病理分析结果显示miR-802与组织分化程度、TNM分期、浸润程度显著相关(P<0.05),RAB23与淋巴结转移、TNM分期及组织分化程度显著相关(P<0.05);Pearson相关性分析显示miR-802与RAB23呈显著负相关(P<0.05);Kaplan-Meier曲线显示miR-802高表达组PFS、OS均显著高于低表达组(P<0.05),RAB23阴性表达组PFS、OS均显著高于阳性表达组(P<0.05)。结论miR-802在食管鳞癌组织中低表达,RAB23高表达,二者呈负相关,且与患者病情进展及预后有关,可作为早期判断食管鳞癌病情以及评定患者预后的指标。
Objective To investigate the expression levels and clinical significances of miR-802 and RAB23 in esophageal squamous cell carcinoma (ESCC).Methods A total of 120 cases of esophageal squamous cell carcinoma specimens in our hospital from March 2014 to February 2015 were selected as the esophageal squamous cell carcinoma group,and the para cancerous normal tissue were selected as the control group (120 cases).The relative expressions of miR-802 and RAB23 mRNA in esophageal squamous cell carcinoma tissues and para cancerous tissues was detected by qRT-PCR.Immunohistochemical method was used to detect the expression of RAB23 protein in esophageal squamous cell tissue.According to the test results,miR-802 was divided into high expression group and low expression group.RAB23 was divided into positive expression group and negative expression group.The relationships between miR-802 and RAB23 expressions with clinicopathological parameters were analyzed.Pearson method was used to analyze the correlation between miR-802 and RAB23.Kaplan-Meier method was used to analyze the survival of patients with esophageal squamous cell carcinoma.The survival differences of different expression groups were detected by Log-Rank.Results Compared with the normal group,the expression level of miR- 802 in esophageal squamous cell carcinoma group was significantly decreased ( P <0.05),while the expression level of RAB23 mRNA increased significantly ( P <0.05).Immunohistochemistry showed that the positive expression of RAB23 protein in esophageal squamous cell carcinoma group was significantly higher than that in normal group ( P <0.05).The results of clinicopathological analysis showed that miR-802 was significantly correlated with the degree of tissue differentiation and degree of invasion ( P <0.05).RAB23 was significantly correlated with lymph node metastasis,TNM staging and the degree of tissue differentiation ( P <0.05).Pearson correlation analysis showed that miR-802 was negatively correlated with RAB23 ( P <0.05).The Kaplan-Meier curv
