民族药用植物走马胎快繁技术 被引量:5

摘要 以走马胎茎段为外植体,研究了不同植物生长调节剂、光照、pH值和添加物对不定芽的诱导、增殖和生根的影响。研究表明:培养基WPM+2. 0 mg/L BAP+0. 2 mg/L NAA+0. 2 mg/L 2,4-D最有利于走马胎茎段腋芽的诱导;培养基WPM+2. 0 mg/L BAP+0. 2 mg/L NAA有利于走马胎腋芽的不定芽诱导和增殖;走马胎腋芽增殖的最佳p H值为6. 0,当培养光照达到100μmol/(m·s)时,最有利于走马胎不定芽生长;培养基中分别加入1 g/L蛋白胨、50 g/L椰汁和50g/L香蕉,均有壮芽的效果,其中香蕉的效果最佳。最佳的生根培养基是MS+2. 0 mg/L IBA+0. 5 mg/L NAA,生根率达93. 3%,根系生长良好,移栽至椰糠∶河沙=2∶1的混合基质中,走马胎生长良好,成活率达95%。 Effects of different plant growth regulators,illumination intensity,pH and additional nutrient on induce,proliferation and rooting of adventitious buds were studied by taking stem segment of Ardisia gigantifolia Stapf as explant. Results showed that the optimal proliferation medium for Ardisia gigantifolia Stapf was WPM with 2. 0 mg/L BAP、0. 2 mg/L NAA and 0. 2 mg/ L 2,4-D. The optimal pH was 6. 0,plantlets grew well when illumination intensity was 100 μmol/( m·s). 1 g/L peptone, 50 g/L coconut juice and 50 g/L banana were good for cultivating strong seedling,and 50 g/L banana was the best. The best rooting medium was MS medium supplemented with 2. 0 mg/L IBA and 0. 5 mg/L NAA,rooting rate reached 93. 3%,Ardisia gigantifolia Stapf grew well with a survival rate of 95% when transplanted into mixed matrix of coconut tree branny: river sand = 2 ∶ 1.
