响应曲面法优化固肾解毒胶囊提取工艺及其对糖尿病肾病大鼠肾功能的影响研究

Optimization of Extraction Process of Gushen Jiedu Capsules by Response Surface Methodology and Its Effects on Renal Function in Diabetic Nephropathy Rats

摘要 目的采用Box-Behnken响应曲面法优化固肾解毒胶囊(Gushen Jiedu Capsules,GSJD)的提取工艺,考察GSJD对糖尿病肾病大鼠肾功能的影响。方法以盐酸小檗碱提取量作为响应值,在单因素实验的基础上采用响应曲面法考察液料比、提取时间、提取次数及其交互作用对盐酸小檗碱提取量的影响并进行验证试验;对大鼠给予高脂饲料合并腹腔注射链脲佐菌素以建立糖尿病肾病大鼠模型,造模成功后的动物给药8周,测定血清生化指标尿素氮并进行肾脏病理组织结构检查。结果GSJD的最佳提取工艺条件为液料比8:1,提取时间2 h,提取次数2次。在此工艺下盐酸小檗碱提取量的预测值为21.09 mg/g,实测值为20.88 mg/g,误差为0.99%;给药8周后,与正常对照组比较,模型组尿素氮水平显著上升,福辛普利组及GSJD高剂量组可显著降低糖尿病肾病大鼠血清尿素氮水平(P<0.05),给药GSJD后对肾脏病理组织结构有不同程度的改善,与福辛普利组结果类似。结论经Box-Behnken响应曲面法优化后的GSJD的提取工艺具有简单方便、可操作性强的优点,其对糖尿病肾病大鼠肾功能有一定的改善作用。 Objective To optimize the extraction process of Gushen Jiedu Capsules(GSJD)by Box-Behnken response surface methodology and investigate the effects of GSJD on renal function in diabetic nephropathy rats.Methods Based on single factor tests,the effects of liquid-solid ratio,extraction time,extraction times and their interaction on the content of berberine hydrochloride in the extract were optimized by response surface methodology.The rats were fed with high-fat diet for 8 weeks and injected intraperitoneally with streptozotocin to induce diabetic nephropathy.Then the rats were treated with or without GSJD by gavage for 8 weeks and the urea nitrogen in serum was examined.In addition,the pathological structure of the kidneys of rats were examined.Results The optimum extraction conditions of GSJD were as follows:the ratio of liquid to material was 8:1,the crude drug was extracted for 2 times,2 h for each time.The predicted extraction content of berberine hydrochloride was 21.09 mg/g,and the measured content was 20.88 mg/g,with an error of 0.99%.After 8 weeks of administration of GSJD,compared with the normal group,the urea nitrogen in the model group increased significantly,while the urea nitrogen in the fosinopril group and GSJD high dose group decreased significantly compared with the model group(P<0.05).The pathological structure of the kidney was improved to varying degrees after administration of GSJD,which was similar to that of the fosinopril group.Conclusion The extraction process of GSJD optimized by Box-Behnken response surface methodology is simple,convenient and operable.Moreover,GSJD can improve the renal function of diabetic nephropathy rats.
