摘要
目的观察乳腺癌患者癌组织中肌苷一磷酸脱氢酶1(IMPDH1)的表达,分析其与乳腺癌临床病理特征的关系。方法随机选取72例乳腺癌患者的癌组织,同时选取距肿瘤边缘大于5 cm的正常乳腺组织作对照,蛋白质印迹法(Western blot)检测比较IMPDH1在乳腺癌及癌旁组织中的表达,免疫组织化学方法检测不同病理特征的乳腺癌组织和癌旁组织中IMPDH1的表达,探讨其与乳腺癌临床病理特征的关系。结果Western blot检测显示IMPDH1在乳腺癌组织的表达水平(4.372±0.326)明显高于癌旁组织(0.649±0.253),差异有统计学意义(P<0.01)。乳腺癌组织的IMPDH1蛋白阳性表达阳性率为79.17%(57/72),高于癌旁正常组织的15.28%(11/72),差异有统计学意义(P<0.01),IMPDH1在乳腺癌组织中的表达与患者年龄、肿瘤大小、月经状态无明显相关(χ^2=1.192,P>0.05;χ^2=2.027,P>0.05;χ^2=0.397,P>0.05),而与乳腺癌腋窝淋巴结转移、细胞核增殖抗原(Ki-67)及TNM分期明显相关(χ^2=8.530,P<0.01;χ^2=4.308,P<0.05;χ^2=4.721,P<0.05)。结论IMPDH1在乳腺癌组织中的表达高于癌旁正常组织,且与乳腺癌腋窝淋巴结转移、Ki-67、TNM分期密切相关,其高表达可能与肿瘤增殖、转移及患者预后有关。
Objective To study the expression level of inosine monophosphate dehydrogenase 1(IMPDH1)in the breast cancer,analyze the correlation between IMPDH1 and clinicopathological characteristics of breast cancer,and explore possible the mechanisms of the involvement of IMPDH1 in the occurrence and development of breast cancer.Methods Seventy-two cases of breast carcinoma and para-carcinoma tissues were collected.Western blotting was used to detect the protein expression of IMPDH1 in breast carcinoma and adjacent tissues.Immunohistochemistry was applied to detect the expression of IMPDH1,and the correlation between IMPDH1 and clinicopathological characteristics of breast cancer was analyzed.Results The positive rate of IMPDH1 expression in breast cancer(79.17%)was significantly higher than in the adjacent tissues(15.28%)(P<0.01).The expression of IMPDH1 was not related to age,size of tumor and menstruation(χ^2=1.192,P>0.05;χ^2=2.027,P>0.05;χ^2=0.397,P>0.05),but to lymph node metastasis,proliferation cell nuclear antigen(Ki-67)and TNM staging(χ^2=8.530,P<0.01;χ^2=4.308,P<0.05;χ^2=4.721,P<0.05).Western blotting showed that the expression of IMPDH1 in breast cancer was significantly higher than in the normal tissue(P<0.01).Conclusion The expression of IMPDH1 in breast cancer is higher than in normal tissues,and is correlated with lymph node metastasis,Ki-67 and TNM staging,suggesting its roles in the breast cancer.
