摘要 探究优化木棉花超氧化物歧化酶(SOD)的提取条件,建立木棉花SOD生产工艺.以木棉花花瓣为原料,采用正交实验优化超声波提取的技术参数,结合丙酮等电点沉淀和葡聚糖凝胶层析提取纯化SOD,经真空冷冻干燥获得SOD成品.超声波提取的最佳工艺为料液比1∶30,功率180w,超声波处理总时间9min,浸提时间8h.沉淀杂蛋白条件为2mol/L的HCl将粗提液调节至pH为5.0,提纯SOD条件为pH4.0丙酮等电点沉淀法,纯化倍数达到5.59,经Sephadex G-75纯化,获得SOD成品,纯化倍数达到18.46.为木棉花SOD的规模化生产提供理论依据. In this paper, it is conducted to optimize the extraction conditions of superoxide dismutase(SOD) from kapok, and to provide an efficient production process for obtaining SOD from kapok. Using kapok petals as raw materials, the best experimental parameters of ultrasonic extraction were optimized by orthogonal test. After that, the extract was precipitated by acetone and purified by dextran gel chromatography. Eventually the SOD product was obtained by vacuum freeze-drying. The optimum technology of ultrasonic extraction is as follows: the ratio of material to liquid is 1:30, the power is 180 w, the total treatment time is 9 min, and the extraction time is 8 h. Then the extract is purified by adjusting the pH to 5.0 to precipitate other proteins with 2 mol/L HCl, and then the pH is adjusted to 4.0 to precipitate SOD with acetone. After purification with Sephadex G-75, the purification multiple of SOD could reach 18.46 and the purified product was obtained. The project’s aim is to provide theoretical underpinning for industrial production of SOD extracted from kapok.
