QCM与光学显微镜联用实时定量研究组胺对人脐静脉内皮细胞黏弹性及铺展面积的影响 被引量:2

摘要 通过QCM技术和光学显微镜技术联用实时监测了不同浓度组胺对人脐静脉内皮细胞(HUVEC)黏弹性与形态的影响。结果表明,组胺刺激细胞后,细胞存储模量、损耗模量和铺展面积均增加,损耗角下降;在25~75μmol·L^(-1)范围内,组胺对HUVEC的作用随着浓度增加而增强;细胞存储模量和损耗模量都与铺展面积之间存在线性关系,通过对比加药前后细胞模量-铺展面积曲线发现,组胺对细胞模量的影响大于对铺展面积的影响。表明QCM用于细胞力学连续定量研究存在可行性,这对进行细胞黏附力学性能定量研究及进一步发展成药物评估工具具有积极意义。 We real-timely monitored the effects of different histamine concentrations on the viscoelasticity and morphology of human umbilical vein endothelial cells(HUVEC) using quartz crystal microbalance(QCM) technique combined with optical microscopy technique.The results show that the cell storage modulus,loss modulus,and spreading area all increase while the loss tangent decreases after the stimulation of histamine.Furthermore,in the range of 25-75 mol·L-1,the effect of histamine on HUVEC increases with increasing of histamine concentration.Moreover,there is a linear relationship between either the cell storage modulus or loss modulus and the spreading area.By comparing the cell modulus-spreading area curve before and after dosing,we find that the effect of histamine on the cell modulus is greater than that on the spreading area.The above results demonstrate the feasibility of QCM for continuous and quantitative study of cell mechanics,which is of positive significance for the quantitative study of cell adhesion mechanical properties and the further development of drug evaluation tools.
