摘要
云龙石斑鱼是云纹石斑鱼(Epinephelus moara♀)和鞍带石斑鱼(Epinephelus lanceolatus?)杂交获得的石斑鱼新品种,具有生长速度快和适温性广的优良性状,其生长速度是母本的1倍以上,具有较高经济效益,已在国内推广养殖.但云龙石斑鱼生长优势的分子机制尚不清楚,本实验选取了相同养殖条件下的云龙石斑鱼和云纹石斑鱼的两个群体各5尾,测量体长、体重等信息,并选取脑、垂体、肝脏和肌肉进行了转录组测序和分析.在脑、垂体、肝脏和肌肉样本中分别得到953、2054、558和1097个基因存在显著的表达水平差异.基于表达模式聚类分析、GO功能显著性富集分析、基因表达通路分析等手段,我们发现,成纤维生长因子,表皮生长因子,血管生成因子等相关基因都明显上调,加快了细胞的增殖分化,并在一定程度上刺激了生长激素的分泌,可能是云龙石斑鱼生长优势的重要贡献因素.研究结果为探讨云龙石斑鱼杂交生长优势形成机制提供了基础数据,为探讨鱼类生长优势的分子机制提供了新思路.
Yunlong grouper,a novel hybrid grouper offspring crossed between Epinephelus moara♀ and Epinephelus lanceolatus,with advantageous traits of fast growth and tolerance to the change of temperature,exhibiting a growth speed twice as fast as its female parent Epinephelus moara,becomes a better cultured species with domestically highly industrialized prospects.In order to understand the molecular mechanism of the growth of Yunlong grouper,transcriptome sequencing and analysis were performed between Yunlong grouper and E.moara.Five individuals of each species from each group were collected and cultured under the same condition.After the body weight and body length were measured,brain,pituitary,liver and muscle were selected for the comparative transcriptomic analysis.953,2 054,558 and 1 097 differentially expressed genes were found in brain,pituitary,liver,and muscle,respectively.Based on expression pattern cluster,GO enrichment and pathway annotation,after furtherly analyzing these differentially expressed genes,we found that the genes involved in fibroblast growth factor,epidermal growth factor,angiogenic factor,receptor tyrosine kinase and other related genes are up-regulated and thus accelerate the proliferation and differentiation of cells,and stimulate the secretion of growth hormone to a certain extent,enabling Yunlong grouper to grow faster.The results shed light on the growth superiority of Yunlong hybrid grouper and provided insights into understating of molecular basis underlying the fish growth.
