青少年期精神分裂症血清蛋白因子水平变化的研究 被引量:5

Study on the level of serum protein factor concentration in adolescent schizophrenia

摘要 目的探讨青少年精神分裂症患者血清中脑源性神经营养因子(BDNF)、神经生长因子(NGF)、髓鞘碱性蛋白(MBP)3种血清蛋白因子治疗前后水平变化及其与精神分裂症临床症状的相关性。方法纳入年龄13~18岁的青少年精神分裂症患者及正常对照组各40例。用酶联免疫吸附测定治疗前后血清中BDNF、NGF、MBP的水平变化;采用PANSS量表评估临床症状的严重程度,并分析上述血清蛋白因子浓度与PANSS评分之间的相关性。结果治疗前,患者组血清BDNF及NGF浓度均低于正常对照组,差异具有统计学意义(P<0.01);MBP的水平高于正常对照组,但差异无统计学意义(P>0.05)。治疗8周后血清中BDNF和NGF浓度跟治疗前相比有所增加,差异有统计学意义(P<0.01);MBP浓度和治疗前相比水平有所下降,但差异无统计学意义(P>0.05)。治疗前NGF的浓度和PANSS总分负相关,BDNF与阴性症状呈负相关,差异有统计学意义(P<0.05)。结论在疾病的早期,神经营养因子BDNF和NGF较神经损伤蛋白MBP更可能的参与了精神分裂症的发生及发展的过程。检测血清BDNF和NGF水平可作为临床观察的指标,将有助于确定精神分裂症临床评估、疗效和预后判断的特异生物学指标,对早期精神分裂症辅助诊断有一定的价值。 Objective To investigate the changes of serum NGF、BDNF and MBP levels pretherapy and posttreatment in the adolescent schizophrenia,and their correlation with clinical symptoms of schizophrenia inpatiens. Methods We recruited 40 adolescent schizophrenia patients(age,13-18 years)and compared them with 40 matched healthy child subjects. The changes of BDNF、NGF and MBP serum levels before and after treatment were measured by using the enzyme linked immunnosorbent assay(ELISA);Psychiatric symptoms before and after treatment were assessed by PANSS scale,and the correlation between the serum protein factor concentration and the PANSS score was studied as well. Result Before treatment,the serum concentrations of BDNF and NGF were lower in the patient group than that in the normal control group,the differences were statistically significant(P<0.01);the levels of MBP in the patient group were higher than that in the normal control group,but the difference was not statistically significant(P>0.05). The levels of BDNF and NGF in patient group were significantly increased after 8 weeks of treatment,the differences were statistically significant(P<0.01),however,the levels of MBP in patient group were decreased after 8 weeks of treatment,but the difference was not statistically significant(P>0.05). There were negative correlation between the PANSS scores and the serum NGF levels before treatment,likewise there were negative correlation in negative symptoms and the serum BDNF levels,and the differences were statistically significant(P<0.05). Conclusion The study suggests that the neurotrophins BDNF and NGF are more likely to participate in the development of disease than the nerve damage protein MBP in the early stages of schizophrenia. The serum levels of BDNF and NGF can be used as a clinical observation indicator,which will help to evaluate clinical characteristics and the treatment effect and the prognosis of schizophrenia.
