摘要
目的在光滑钛表面构建载锶纳米管样结构,评价其对SD大鼠骨髓干细胞成骨分化的影响。方法采用阳极氧化、水热处理的方式在钛片表面形成纳米管、载锶纳米管样结构,通过扫描电镜、能谱分析检测其微观形貌和元素组成,通过细胞形态、黏附、增殖和基因、蛋白表达检测评价其体外生物学功效。结果氢氟酸溶液阳极氧化、氢氟酸溶液阳极氧化加含锶溶液水热处理可以在钛片表面形成纳米管、载锶纳米管结构,不同的表面形貌会影响细胞的形态、黏附和增殖功能,载锶纳米管能显著提高成骨分化相关基因和蛋白的表达水平(P<0.01)。结论载锶纳米管结构可在体外促进SD大鼠骨髓间充质干细胞的成骨分化,上调其成骨相关基因的表达。
Objective To construct strontium loaded nanotubes on titanium sheets and to evaluate its effect on osteogenic differentiation of rat mesenchymal stem cells. Methods Anodic oxidation and hydrothermal treatment were used to form nanotubes and strontium loaded nanotubes on the surface of titanium. The morphology and elemental composition of the nanotubes were examined by scanning electron microscope and energy dispersive analysis. The cell morphology, adhesion, proliferation, expression of genes and proteins were detected in vitro. Results Anodic oxidation and hydrothermal treatment could form nanotubes and strontium loaded nanotubes on the surface of titanium sheet. Different surface structure affected the morphology, adhesion and proliferation of cells, and the strontium loaded nanotubes could significantly up-regulate the expression of osteogenic genes and proteins ( P <0.01). Conclusion Strontium loaded nanotubes on titanium sheets could promote osteogenic differentiation of SD rat bone marrow mesenchymal stem cells and up-regulate the expression of osteogenic genes.
