摘要
为了比较VITKE 2 COMAPCT、 VITEK MS质谱、 16s rDNA基因测序3种方法鉴定洋葱伯克霍尔德菌群的异同,挑选15株从米面制品、淀粉及其制品中分离得到的洋葱伯克霍尔德菌群的菌,分别用VITEK 2 COMPACT、 VITEK MS质谱和16s rDNA基因测序进行鉴定。结果显示, VITEK 2COMPACT只能鉴定到群水平,可信度88%—99%; VITEK MS质谱可鉴定到种水平,鉴定置信度99%;16s rDNA基因测序可鉴定到种水平,序列同源性能达到99%以上,但每株菌16s序列能匹配到2种以上洋葱伯克霍尔德菌群里的菌。所以, 3种鉴定方法中, VITEK MS质谱法可鉴定出洋葱伯克霍尔德菌群里的不同种,该方法结果准确,时效性高,操作步骤简单。
In order to compare the differences and similarities among VITKE 2 COMAPCT, VITEK MS and 16s rDNA gene sequencing in the identification of Burkholderia cepacia, 15 strains of Burkholderia cepacia isolated from rice noodles, starches and their products are selected and identified by VITEK 2 COMPACT, VITEK MS and 16s rDNA gene sequencing, respectively. The results show that VITEK 2 COMPACT can only identify the population level with 88%-99% reliability;VITEK MS can identify species level with 99% confidence;16s rDNA gene sequencing can identify species level with 99% homology, but 16s sequence of each strain can match more than two species of Burkholderia cepacia. So, among the three identification methods, VITEK MS can identify different species of Burkholderia cepacia group, and it has the advantages of accurate results, high timeliness and simple operation steps.
