摘要
目的建立市售菟丝子饮片的UPLC指纹图谱,考察其质量的一致性,为其质量评价提供参考。方法采用Agilent SB-C_(18)(2.1 mm×100 mm,1.8μm)色谱柱;以乙腈-0.2%甲酸水溶液为流动相,梯度洗脱;流速:0.4 mL/min;柱温:35℃;检测波长:采用切换法(1~5 min,325 nm;5~12 min,260 nm;12~30 min,360 nm),对所得各指纹图谱进行相似度计算、聚类分析和主成分分析。结果建立了菟丝子饮片的UPLC指纹图谱,标定共有峰10个,并鉴定出其中5个色谱峰的化学成分,分别为绿原酸、金丝桃苷、异槲皮苷、紫云英苷、山柰酚。102家生产企业112批次样品中,11批次样品相似度范围在0.659~0.930之间,其余101批次样品相似度均在0.950以上,且能较好的聚为一类。结论大部分市售菟丝子饮片质量一致性较好,也有部分存在一定差异。该方法简便、可靠,可用于菟丝子的质量评价。
Objective To establish a UPLC fingerprint of Cuscutae Semen (CS),and to investigate the consistency of its quality for providing reference for its quality control.Methods The UPLC separation was performed on an Agilent SB-C 18 reversed-phase column (2.1 mm×100 mm,1.8 μm) in gradient mode of acetonitrile-0.2% formic acid with PDA detection at 1-5 min,325 nm;5-12 min,260 nm;12-30 min, 360 nm.The column temperature was 35 ℃,and the flow rate was 0.4 mL/min.The fingerprints were evaluated by similarity evaluation,cluster analysis and principal components analysis.Results The UPLC fingerprint of CS was developed.There were ten common characteristic peaks in the UPLC fingerprints and five of which (chlorogenic acid,hyperin,isoquercitrin,astragalin,kaempferide) were identified.In the 112 batches samples from 102 manufacturers,the similarities of eleven batches were between 0.659-0.930,and the other 101 batches had similarities above 0.950,which could be clustered into one group.Conclusion The quality consistency of CS on the market was good from the most manufacturers,while there are also significant differences in some manufacturers.This simple and reliable method can be used for the quality control of CS.
